中文摘要:【目的】研究金钱鱼(Scatophagus argus)全基因组微卫星分布特征,筛选高多态性微卫星(SSR)标记。【方法】利用MISA软件对金钱鱼基因组中微卫星进行筛选并分析分布特征,利用聚丙烯酰胺凝胶电泳和毛细管电泳筛选高多态性位点。【结果与结论】金钱鱼基因组总共挖掘390967个SSR位点,相对丰度为653个/Mb。二碱基重复类型最多(52.64%),微卫星标记整体偏向A+T碱基组合。筛选出23对多态性标记位点,其中7对的等位基因数为4~11,期望杂合度为0.431~0.859,观测杂合度0.417~0.861,多态性含量为0.389~0.830。经Bonferroni校正后,5个位点符合哈德温伯格平衡,且位点间不存在连锁不平衡现象。

外文摘要:【Objective】To develop highly polymorphic SSR markers and analyze the whole genome microsatellite distribution patterns in Scatophagus argus.【Methods】The MIcro SAtellite(MISA)identification software was employed in data mining and isolate the microsatellites loci in S.argus.Microsatellite polymorphism was explored by capillary electrophoresis and polyacrylamide gel electrophoresis.【Result and Conclusion】A total of 390967 microsatellites loci were identified in the genome of S.argus,with a relative abundance of 653 per Mb.Among the 1 to 6 different base repeat types of microsatellites,the number of two base repeats was the highest,and the A+T were the most frequent microsatellite classes.A total of 23 pairs of polymorphic markers were identified from the screening,and capillary electrophoresis was performed on 7 of them.The number of alleles per locus ranged from 4 to 11.The expected and observed heterozygosity ranged from 0.431 to 0.859 and from 0.417 to 0.861,respectively.The polymorphism information content ranged from 0.389 to 0.830.After Bonferroni correction,five loci were consistent with the Hardy-Weinberg equilibrium and showed no linkage disequilibrium.
