中文摘要:以广东地方黄羽肉鸡麒麟鸡、怀乡鸡为研究对象,采用高效液相色谱法测定不同时间鸡肉肌苷酸(IMP)含量,利用实时荧光定量PCR法比较影响肌苷酸含量的GPAT和PURH基因在不同时间各组织中的表达水平,并研究2种基因在不同鸡中的表达量与肌苷酸含量的相关性。结果表明:怀乡鸡鸡肉中肌苷酸含量最高时为16周龄,麒麟鸡则为14周龄。GPAT、PURH基因在麒麟鸡各组织表达量随日龄增加均呈下降趋势,而怀乡鸡则呈先升后降的变化。麒麟鸡和怀乡鸡GPAT基因3个时间点中肝脏的表达量最高,而PURH基因3个时间点胸肌的表达量最高。GPAT在麒麟鸡和怀乡鸡肝脏的表达量与腿肌肌苷酸含量呈极显著负相关,PURH在怀乡鸡胸肌的表达量与胸肌肌苷酸含量呈显著负相关。

外文摘要:In this study, the Guangdong Local breeds Yellow Broilers of Kirin chicken and Huaixiang chicken were selected. The inosine monophosphate (IMP) content in muscle of chicken of different time was detected by HPLC. The GPAT and PURH gene expression levels in some tissue which can influence the IMP content were exam- ined by Real-time quantitative PCR. A correlation analysis was made on the expression levels of two genes in different breeds and TMP content. The results showed that the IMP highest content in muscle of Huaixiang chicken was 16 weeks, the IMP highest content in muscle of Kirin chicken was 14 weeks. PURH and GPAT gene expression in Kirin chicken organizations were decreased with the increase of age, while Huaixiang chicken is increased first then de- creased. GPAT gene expression of Kirin chicken and Huaixiang chicken showed the highest in liver at all three time points. While PURH gene expression at all three time points in chest muscle is the highest. There is a significantly negative correlation between IMP content in leg muscle and GPAT gene expression in liver of Kirin chicken and Huaix- iang chicken. PURH gene expression in Chest muscle of Huaixiang chicken and IMP content in chest muscle of Hua- ixiang chicken was negatively correlated.
