中文摘要:目的盐酸小檗碱体外抗肿瘤等作用活性显著,目前针对其从体外过渡到体内的研究偶见报道,文中研究通过半体内研究方法,比较盐酸小檗碱含药血清、血浆对人鼻咽癌CNE-1细胞的生长抑制及诱导凋亡作用。方法新西兰白兔灌胃给药,分离血清、血浆后,56℃灭活30 min,制成含药血清、血浆,采用反向高效液相色谱法(rerersed phase high performancel iquid chromatography,RP-HPLC)分析盐酸小檗碱含量,采用MTT法比较不同作用时间和剂量对鼻咽癌CNE-1细胞增殖的抑制作用,用Annexin v-FITC流式细胞术观察诱导细胞凋亡情况。结果盐酸小檗碱含药血清、血浆均能够抑制CNE-1细胞增殖,含药血浆的抑制作用强于含药血清,且含药血浆能显著诱导CNE-1细胞凋亡。结论凡是使用血清与血浆均可的实验,应当优先选用含药血浆。

外文摘要:Objective Berberine hydrochloride (BH) has an obvious anti-tumor activity in vitro, and studies on its effect are reportedly from in vitro to in vivo. This article aims to compare the inhibiting and apoptosis-inducing effects of BH in the serum and plasma on nasopharyngeal carcinoma CNE-I cells using semi-in vivo research methods. Methods New Zealand rabbits were given BH by gavage, and then the serum and plasma were separated from the animals and inactivated in 56~C water for 30 minutes. The con- tents of BH in the serum (SBH) and plasma (PBH) were determined by RP-HPLC, the effects SBH and PBH on the proliferation of the CNE-1 cells evaluated by MTr assay, and the apoptosis of the cells detected using Annexin V-FITC flow cytometry. Results Both SBH and PBH suppressed the growth of the CNE-1 cell line, and PBH exhibited an even better inhibitory effect than SBH. Be- sides, PBH induced a significant apoptosis of the CNE-1 cells. Conclusion SBH and PBH can inhibit the proliferation and induce the apoptosis of CNE-1 cells, and PBH has even better effects than SBH.
