中文摘要:酚类物质是桑叶的重要活性物质之一,利用常规乙醇-水溶液提取工艺制备的桑叶粗提物中酚类物质占比较低且大分子杂质多,不利于桑叶酚类单体组成研究及后续产品研发。以酚类物质含量较高的桑叶为提取原料,开展桑叶多酚粗提物的富集、纯化工艺研究。以桑叶多酚富集效果为考察指标,筛选出吸附和解吸能力最佳的D101型树脂,并在此基础上进一步优化分离纯化条件,获得最佳吸附工艺参数为:进料液多酚质量浓度1 mg/m L、p H 3. 0,上样流速1 m L/min,上样量11倍柱床体积(BV)。获得的最佳解吸工艺参数为:1 BV 10%乙醇去除多糖、还原糖、水溶性蛋白等杂质,3 BV 30%乙醇和3 BV 50%乙醇以1 m L/min的流速洗脱收集。纯化后,经HPLC鉴定和分析,桑叶多酚主要含有绿原酸、芦丁等11种酚类单体,其中绿原酸、芦丁、金丝桃苷、安息香酸4种单体含量较高,占可检测总酚类单体含量的93. 6%。纯化后,总酚质量比由纯化前的198. 8mg/g提高到752. 9 mg/g,绿原酸、芦丁、金丝桃苷、安息香酸分别由纯化前的99. 3、39. 1、12. 7、35. 0 mg/g提高到纯化后的408. 3、112. 0、58. 1、122. 0 mg/g。研究结果可为桑叶酚类单体组成研究及桑叶多酚功能食品研发提供参考。
外文摘要:Polyphenols are one kind of important functional components in mulberry leaves. The crude extract from mulberry leaves by conventional ethanol-water solution extraction process accounts for a relatively low percentage of phenolic substances and a large number of macromolecular impurities,which is not conducive to the research on composition of phenolic monomer in mulberry leaves and the following product developing process. In this study,mulberry leaves with high content of phenolic compounds were used as materials to study the enrichment and purification process of polyphenol crude extract from mulberry leaves. Based on enrichment effect of macroporous adsorption resin,taking the adsorption and resolution ability of mulberry polyphenols as index,D101 resin with the best performance was selected for the process optimization. The optimized adsorption conditions were: polyphenol concentration in loading solution of 1 mg/m L,pH of3. 0,flow rate of 1 m L/min and the load volume of 11-fold bed volume( BV). The optimized desorption conditions were:after adsorption,1 BV of 10% ethanol was applied to the column to remove the impurities including polysaccharides,reducing sugars and water soluble proteins; Polyphenols were collected with elution of 3 BV 30% ethanol and 3 BV 50%ethanol at a flow rate of 1 m L/min. After purification,according to HPLC identification and analysis,polyphenols in mulberry leaves mainly contained 11 phenolic monomers such as chlorogenic acid and rutin etc. Four kinds of phenolic monomers,namely chlorogenic acid,rutin,hyperoside and benzoic acid,account for about 93. 6% of the total phenolic monomers detected. After purification,total phenol content in the purified product increased from 198. 8 mg/g to 752. 9 mg/g,contents of chlorogenic acid,rutin,hyperoside and benzoic acid increased from 99. 3,39. 1,12. 7 and 35. 0 mg/g to 408. 3,112. 0,58. 1 and 122. 0 mg/g,respectively. This study provides useful references for further research on mulberry phenolic monomer composition and development of functional food containing mulberry polyphenols.
