中文摘要:环境热应激是制约我国南方地区家禽养殖业的一个重要因素,在饲料中添加姜黄素可以有效提高应激家禽的生产性能。circRNA是新发现的一类非编码RNA,通过调控miRNA及其介导的基因沉默在抑制细胞蛋白应激变性中发挥重要作用。本试验对比分析在夏季高温环境下,基础日粮中添加姜黄素缓解成年蛋鸡热应激过程中卵巢组织circRNAs差异表达谱。90只26周龄成年蛋鸡随机分为常温对照、热应激和姜黄素保护3组,分别饲喂姜黄素含量为0或200 mg/kg的日粮,30 d后每3组分别随机选取3只,采集卵巢组织制备HE染色组织切片并测序分析circRNAs差异表达谱。结果显示,相比对照组和姜黄素保护组,高温组切片中卵泡的内容物增多、着色加深,卵泡壁增厚。卵巢组织测序数据统计分析发现,与常温对照组、姜黄素保护组相比,热应激组中分别有21和27个circRNAs表达水平差异极显著(P<0.01);而3组间表达差异显著(P<0.05)的circRNAs共有66个。对3组间表达差异circRNAs的基因功能分析发现,其中与热应激-抗热应激过程密切相关(P<0.05)的调控通路有细胞骨架调节和黏着连接、Nod样受体信号通路、ErbB和FoxO信号通路,并进一步构建circRNAs-miRNAs靶向调控关系图。以上研究为解析姜黄素缓解蛋鸡卵巢热应激,提高蛋鸡生产性能提供重要试验数据。
外文摘要:The heat stress caused by continuous high temperature weather has become an important restricting factor in the poultry industry.Curcumin added in the feed can enhance animal organism oxidation resistance and effectively improve poultry production performance in the stress condition.CircRNA as a new kind of non-coding RNA,played a important role in inhibition of cell protein denaturation under stress by mediating gene silencing.In this study,the differential expression profile of circRNAs in ovarian tissue of laying hens were investigated in normal temperature group,high temperature group and curcumin protection group under the environment of high temperature in the summer.Ninty adult laying hens with 26 weeks old were randomly divided into three groups,fed a diet with curcumin content of 0 or 200 mg/kg for 30 d respectively.Then the ovarian tissue of 9 hens from the three groups of 3 each were collected to prepare HE stained tissue sections and analyzed differential expression profiles of circRNA by deep sequencing.In the section of ovarian tissue,compared with the control group and curcumin protection group,the contents of follicles were increased,the color was deepened,and the follicular wall was thickened in the high-temperature group.The statistical analysis of sequencing data found that there were 21 and 27 circRNAs with the extremely significant difference expression level(P<0.01)in high temperature group compared with the normal control group and curcumin protection group in ovarian tissue,respectively.The number of circRNAs with significant difference expression level(P<0.05)among the three groups was a total of 66;the function analysis of difference expressed circRNAs found that the regulation pathways closely related to the heat stress and anti-heat stress process(P<0.05)were cytoskeletal regulation and adhesion connections,Nod like receptor signaling pathways,ErbB,FoxO signaling pathways,and the targeted regulatory diagram of circRNAs-miRNAs were also constructed.This research provided important experimental data for the further function analysis of curcumin in relieving ovarian heat stress and improving the production performance of laying hens.
