中文摘要：转化生长因子β1(transforming growth factor beta1,TGF-β1)基因是本课题组前期在高产和低产雷州黑鸭群体中通过转录组学测序分析筛选出的可能对雷州黑鸭繁殖性能有重要调控作用的候选基因。为进一步探究TGF-β1基因的遗传多态性,分析其产蛋性能的相关的遗传分子标记,本试验采用PCR扩增技术和直接测序法,检测100只雷州黑鸭群体中的TGF-β1基因的SNP位点,使用SPSS 26.0软件分析TGF-β1基因的遗传多态性位点基因型与雷州黑鸭产蛋性能的相关性。结果显示,在TGF-β1基因的第6个内含子中发现3个SNPs位点与雷州黑鸭的开产蛋重显著相关,分别为I6-2188 C>T、I6-2220 T>C和I6-2235 A>C;I6-2188 C>T位点与开产日龄显著相关;使用Haploview软件对SNPs位点进行连锁分析发现,H1H1单倍型的开产蛋重显著高于H2H2单倍型。综上所述,TGF-β1基因的3个SNPs位点可作为雷州黑鸭繁殖性能的遗传标记。

外文摘要：Transforming growth factor beta1 gene is a candidate gene that may have an important regulatory role in the reproductive performance of Leizhou black ducks.TGF-β1 was screened out by transcriptome sequencing analysis of high-yield and low-yield Leizhou black duck populations.This study investigated the genetic polymorphism of TGF-β1 gene in Leizhou black duck.The genetic molecular markers associated with laying performance of Leizhou black ducks were analyzed.In this experiment,genomic DNA of 100 Leizhou black ducks was extracted to identify single nucleotide of the TGF-βgene.Polymerase chain reaction techniques were used to obtain the TGF-βgene and the amplification products of the TGF-βgene were analyzed by direct sequencing.The correlation between the genetic polymorphic and genotypes of the TGF-β1 gene and laying performance of Leizhou black ducks were analyzed by SPSS 26.0 software.The results showed that three SNPs,I6-2188 C>T,I6-2220 T>C and I6-2235 A>C,were found in intron 6 of the TGF-β1 gene and were significantly correlated with egg weight of Leizhou black ducks;I6-2188 C>T was significantly correlated with first egg age.The linkage analysis of SNPs loci was performed by using Haploview software,it revealed that egg weight of H1H1 haplotype was significantly higher than that of H2H2 haplotype.In conclusion,the three SNPs loci of the TGF-β1 gene can be used as molecular markers for reproductive performance of Leizhou black ducks.
