中文摘要:目的:研究毛蚶糖胺聚糖(SSG)对高脂血症小鼠的降血脂作用以及对动物细胞内胆固醇生物合成的影响。方法:试验持续4周,用试剂盒测定血清中TC、TG、LDL-C、HDL-C的含量;取小鼠肝脏,石蜡切片,光镜观察小鼠肝脏细胞;通过体外培养中国仓鼠卵细胞(CHO),用两性霉素B-细胞模型法观察SSG对细胞内胆固醇生物合成的抑制作用,用酶联免疫法检测CHO细胞的存活率。结果:SSG中、高剂量能显著降低高脂血症小鼠血清中TC、TG、LDL-C含量和AI(LDL-C/HDL-C)值(P<0.05),使高脂血症小鼠的脂肪肝有明显的减轻;毛蚶SSG2-3-2、SSG2-3-1有保护CHO细胞免受两性霉素B损伤作用(P<0.01)。结论:毛蚶糖胺聚糖对实验性高血脂小鼠具有显著的降血脂作用和预防脂肪肝作用,其作用机制与细胞内胆固醇的生物合成抑制有关。
外文摘要:Objective: The reducing blood lipid of hyperlipidemia mice and effect on cholesterol biosynthesis in animal cells of glycosaminoglycan from Scapharca subcrenata (SSG) were conducted in this study.Methods: The experiment was lasted for 4weeks.The cholesterol(TC),Triglyceride (TG),low-density cholesterol (LDL-C) and high-density cholesterol (HDL-C) in the serum were tested using kit and then the AI(LDL-C/HDL-C) value was got from the formula.The liver cells were observed by paraffin-sections and optical microscope.The amphoterin B cell model was set up to detect the inhibition effect of SSG on the cholesterol biosynthesis in cultured Chinese hamster oocytes(CHO) cells.The survival rate of CHO cells were detected by enzyme linked immunosorbent assay.Results: The medium and high dose groups could decrease the contents of TC,TG,LDL-C and AI(LDL-C/HDL-C) value(P0.05).The fatty liver was prevented and cured effectively too.The survival rate of CHO cells which were pre-incubated with SSG2-3-1,SSG2-3-2 increased,and showed significant difference(P0.01) compared with the control group.This results indicated SSG can protect CHO cells from damage in amphotericin B.Conclusions: SSG can significantly reduce the blood lipids of hyperlipidemia mice and prevent the formation of fatty liver.It’s mechanism may be related to the inhibition of cholesterol biosynthesis in cells.
