中文摘要:雌激素受体(estrogen receptor,ER)是核受体超家族的重要成员,在生殖系统发育、骨骼形成、神经系统发育过程中发挥重要作用。为探究ER在马氏珠母贝性腺发育中的作用,本研究采用RACE技术获得马氏珠母贝雌激素受体(PmER)的c DNA序列全长并检测其在不同组织的表达模式。结果表明:PmER序列全长3517 bp,其中5’UTR为382 bp,3’UTR为1809 bp,开放阅读框(ORF)为1326 bp,编码441个氨基酸。预测其分子量(MW)为49.40 kD,理论等电点(PI)为8.96。SMART软件分析PmER氨基酸序列具有典型的DNA结合结构域(DBD)和配体结合结构域(LBD)。多序列比对发现PmER与其它物种的同源性较高。qPCR结果表明,PmER在性腺显著高表达,其次是外套膜边缘区和中央区。综上所述,PmER可能参与马氏珠母贝性腺发育过程,为进一步探究PmER在马氏珠母贝性腺发育中的作用提供基础资料。

外文摘要:Estrogen receptor(ER)is a key member of nuclear receptor superfamily and plays an important role in reproductive system development,bone formation and nervous system development.To explore the role of ER in gonadal development of Pinctada fucata martensii,full length cDNA sequence of estrogen receptor gene from Pinctada fucata martensii(PmER)was obtained by RACE and we also detected its expression patterns in different tissues.The results showed that the full length cDNA Sequence of PmER was 3517 bp,of which 5’UTR was 382 bp,3’UTR was 1809 bp,ORF was 1326 bp,encoding 441 amino acids.The molecular weight(MW)and theoretical isoelectric point(pI)were predicted to be 49.40 kD and 8.96 respectively.SMART software analysis of PmER amino acid sequence has typical DNA binding domain(DBD)and ligand binding domain(LBD).Multiple sequence alignment showed that PmER had high homology with other species.The results of qPCR showed that PmER was highly expressed in gonads,followed by marginal and central areas of mantle.In conclusion,PmER may be involved in the gonadal development of Piuctada fucata martensii,providing basic data for further exploring the role of PmER in the gonadal development of Piuctada fucata martensii.
