中文摘要:采用酸碱法(pH-shifting)提取罗非鱼下脚料蛋白,主要探讨提取的pH对提取蛋白的得率、分离蛋白的基本成分、溶解性、乳化性和分子量分布的影响。结果表明:在料液比1∶9(g/g)、pH 2.0、pH 3.0以及pH 12.0、pH 13.0条件下溶解,pH 5.5条件下沉淀,提取蛋白得率达52%以上。冷冻干燥制备罗非鱼下脚料分离蛋白粉,其蛋白含量高于85%(干基)。功能特性的研究显示,在pH12.0条件下提取的分离蛋白功能特性最好,其溶解度和乳化活性指数分别为60.80%和42.07 m2/g。SDS-PAGE电泳分析显示,不同pH条件下提取的罗非鱼下脚料蛋白的分子量分布差别不大,主要显示分子量为200 ku和44.3 ku的蛋白条带,分别是肌原纤维蛋白组分中的肌球蛋白和肌动蛋白,另有部分蛋白条带在14.30 ku,属于小分子的肌浆蛋白。
外文摘要:Fish protein isolates was prepared from Tilapia by-products by pH-shifting in this study.Effect of pH condition in the extraction processing on yield of protein,nutritional components,solubility,emulsifying properties and molecular weight distribution of protein isolates was determined.Results showed that protein solution was obtained from Tilapia by-products at 1∶9(g∶g) of concentration of substrate,pH 2.0,3.0 and pH 12.0,13.0,and then was precipitated at pH 5.5,in which protein yield was more than 52 %.Protein isolates powder was obtained by freeze-drying,and content of crude protein in dry basis was more than 85 %.Studies of functional properties showed that protein isolates extracted at pH condition of 12.0 had the best solubility and emulsifying activity,and solubility and emulsifying activity index were 60.80 % and 42.07 m2/g.Studies of SDS-PAGE showed that several protein isolates from different extracting conditions had similar and complex molecular weight distribution,and protein strips displayed mainly at 200 ku and 44.3 ku,which these were myosin and actin.Water-soluble micromolecule sarcoplasmic protein strip distributed at 14.30 ku.
