中文摘要:采用Caco-2单层细胞模型体外模拟马氏珠母贝高F值寡肽(PHFP)小肠吸收,研究结果显示:无论是AP面→BL面还是BL面→AP面,PHFP的吸收量均呈增加趋势。添加P-糖蛋白的专属抑制剂维拉帕米后,AP→BL的渗透系数由原来的6.83×10-7cm/min提高到8.16×10-7cm/min,而BL→AP渗透系数由8.84×10-7cm/min降低到2.20 10-7cm/min,说明PHFP溶液在Caco-2细胞中的转运存在外排泵的作用。并且,PHFP在Caco-2细胞转运过程中被细胞中所存在的代谢酶所代谢,而添加维拉帕米后,PHFP的代谢量有所降低。
外文摘要:Based on the sequence alignment of Bacillus amyloliquefaciens,two pairs of primers were designed and synthesized in this study.Using the total DNA of Bacillus amyloliquefaciens as template,the gene of β-1,3-1,4-glucanase(bgl) was amplified by PCR,which has about 750 bp in length.The gene was cloned into pTG19-T Easy vector,and the recombinant plasmid was identified by PCR,restriction enzyme analysis and sequencing.The homology analysis revealed that the nucleotide sequences of the cloned β-1,3-1,4-glucanase gene similar to the B.amyloliquefacines(M15674),B.subtilis(D00518) and B.licheniformis(AY365256)were 99%,95% and 94%,respectively.
