中文摘要:【目的】为获得可用于动物益生菌制剂的潜在菌种资源,本研究从青海牧民传统酸奶中分离乳酸菌,重点鉴定其中的鼠李糖乳酪杆菌,并对其安全性、黏附性及抗菌性等关键益生特性进行评估。【方法】采用MRS琼脂培养基,在厌氧条件下对青海牧民传统酸奶中的乳酸菌进行分离纯化;通过菌落形态学观察结合16S rRNA基因测序比对进行菌种鉴定;采用绵羊血琼脂平板法测定酸奶样品中鼠李糖乳酪杆菌的溶血活性;通过蛋白酶与淀粉酶分泌试验分析代谢功能;采用自凝集试验、黏附试验及微生物黏附碳氢化合物法表征菌体表面的黏附潜力。采用纸片扩散法检测分离菌株对10种抗菌药物的敏感性,并运用琼脂扩散法评估其对多种致病菌的抑制能力。【结果】从酸奶样品中成功分离得到3株鼠李糖乳酪杆菌:LGG3、TF1、BAMN3,所有菌株均无溶血活性,且均能分泌蛋白酶与淀粉酶。菌体表面特性分析显示,LGG3菌株自凝集率显著高于BAMN3菌株(P<0.05),疏水率和黏附率均显著优于TF1和BAMN3菌株(P<0.05)。抗菌药物敏感性分析表明,3株菌均对头孢噻肟、卡那霉素、庆大霉素表现耐药,而对四环素敏感,对氨苄西林、氯霉素和克林霉素敏感或中敏。在抑菌活性方面,LGG3菌株对革兰阴性和革兰阳性致病菌均表现出广谱抑制效果。【结论】鼠李糖乳酪杆菌LGG3具备良好的体外益生特性,包括有效的表面黏附能力及显著的抗菌活性等,可作为开发动物益生菌剂的潜在候选菌株。
外文摘要:【Objective】To obtain potential bacterial resources usable for animal probiotic preparations,this study isolated lactic acid bacteria from the traditional yogurt made by herdsmen in Qinghai,focused on identifying Lacticaseibacillus rhamnosus(L.rhamnosus)among them,and evaluated its key probiotic properties including safety,adhesiveness,and antibacterial activity.【Method】Lactic acid bacteria were isolated and purified from the traditional yogurt made by herdsmen in Qinghai using MRS agar medium under anaerobic conditions.Strain identification was conducted through colony morphological observation combined with 16S rRNA gene sequence alignment.The hemolytic activity of L.rhamnosus in yogurt samples was determined using the sheep blood agar plate method,and the metabolic functions were analyzed through protease and amylase secretion experiments.The adhesion potential of the bacterial cell surface was characterized using selfagglutination assay,adhesion experiments,and the microbial adhesion to hydrocarbons(MATH)method.The sensitivity to 10 types of antimicrobials was detected by disk diffusion method,and the inhibitory ability against pathogenic bacteria was evaluated by agar diffusion method.【Result】Three strains(LGG3,TF1,and BAMN3)of L.rhamnosus were successfully isolated from yogurt samples,all strains showed no hemolytic activity and were capable of secreting protease and amylase.Analysis of the bacterial cell surface characteristics revealed that the self-agglutination rate of LGG3 strain was significantly higher than that of BAMN3 strain(P<0.05),and the hydrophobicity and adhesion rates were significantly higher than those of TF1 and BAMN3 strains(P<0.05).Antimicrobial sensitivity analysis results showed that all three strains were resistant to cefotaxime,kanamycin and gentamicin,sensitive to tetracyclines,and sensitive or moderately sensitive to ampicillin,chloramphenicol and clindamycin.In terms of antibacterial activity,LGG3 strain exhibited a broad-spectrum inhibitory effect against both Gram-negative and Gram-positive pathogens.【Conclusion】L.rhamnosus LGG3 exhibited excellent probiotic properties in vitro,including effective surface adhesion capability and significant antibacterial activity,it could be used as potential candidate strain for the development of animal probiotic preparations.
