外文摘要:The tire track eel (Mastacembelus armatus) is a commercially and ornamentally valuable teleost species increasingly threatened by Aeromonas hydrophila infection, yet the cellular basis of its antibacterial immunity remains unexplored. Here, we performed single-nucleus RNA sequencing (snRNA-seq) of the M. armatus head kidney following A. hydrophila challenge. We captured 11,399 high-quality nuclei and constructed a comprehensive cell atlas comprising both immune and non-immune cell populations. Notably, we found no transcriptionally discrete non-specific cytotoxic cell (NCC) subpopulation: NCCRP1-positive cells were scattered across multiple lineages, and canonical tilapia NCC markers were diffusely expressed rather than confined to a distinct cluster. Following infection, monocytes/macrophages (Mos/M phi s) exhibited the most extensive transcriptional remodeling and differentiation, characterized by pronounced activation of the lysosome pathway and cathepsin upregulation. In contrast, granulocytes and renal interstitial cells showed abundant differentially expressed genes but minimal trajectory remodeling, while glomerular cells displayed limited phagosome-pathway-associated differentiation. A shared mitochondrial and oxidative metabolic response was observed across diverse cell types. These findings reveal the absence of a discrete NCC lineage in M. armatus and establish Mo/M phi as the principal responders to A. hydrophila infection, providing a species-specific resource for future vaccine design and disease management.
