中文摘要:【目的】优化选择性脂肪酶水解金枪鱼油制备改性水解甘油酯的工艺参数,分析脂质改性前后甘油酯的脂质组分差异。【方法】综合考虑水解度、水解速度和水解甘油酯中ω-3 PUFAs含量,筛选出能选择性富集ω-3 PUFAs的两种脂肪酶,并分别由单因素试验和正交试验优化两种脂肪酶水解鱼油的工艺参数。【结果与结论】筛选出能富集ω-3 PUFAs、改善脂肪酸组成的脂肪酶Lipozyme?TL 100L(TL 100)和NovoCoADL(ADL),其中TL 100的适宜水解条件为缓冲液pH值7.0,温度40℃,水油质量比2∶1;ADL的适宜水解条件为缓冲液pH值7.0,温度45℃,水油质量比2∶1。脂肪酶TL 100和ADL在该适宜条件下水解金枪鱼油,在30 h的水解度分别为(54.37±3.20)%和(45.94±1.50)%,所得水解甘油酯中EPA和DHA总比例分别为(54.89±0.64)%和(51.01±2.40)%,ADL催化水解能显著降低水解甘油酯中的棕榈酸含量,增加油酸含量。选择性脂肪酶水解金枪鱼油能显著提高ω-3 PUFAs含量,改善脂质的脂肪酸分布,制备脂质改性水解甘油酯,但所得甘油酯氧化稳定性显著降低(P<0.05)。
外文摘要:【Objective】The process parameters of preparing lipid-modified acylglycerol from hydrolyzed tuna oil by selective lipase was optimized and the lipid components in acylglycerol before and after lipid modification was analyzed.【Method】Considering the degree of hydrolysis,hydrolysis speed and the ω-3 PUFAs content in concentrated acylglycerol.The two lipases that could selectively enrichω-3 PUFAs were screened and the process parameters of tuna oil hydrolyzed by selective lipase were optimized by single factor test or orthogonal test.【Result and Conclusion】Lipase TL 100 and ADL were used to enrichω-3 PUFAs and to prepare hydrolyzed tuna oil acylglycerol with improved fatty acid profile.The optimal hydrolysis conditions of TL 100 were:buffer pH 7.0,temperature 40℃,and ratio of water to oil was 2∶1;the optimal hydrolysis conditions of ADL were at pH 7.0,temperature 45℃,and ratio of water to oil was 2∶1.Under these conditions,the hydrolysis degree of lipase TL 100 and ADL at 30 h were(54.4±3.2)%and(45.9±1.5)%,respectively.The total contents of EPA and DHA in the concentrated acylglycerol were(54.9±0.6)%and(51.0±2.4)%,respectively.Furthermore,the hydrolysis by ADL can significantly reduce palmitic acid content and increase oleic acid content.Hydrolysis of tuna oil by selective lipases can modify the fatty acids distribution to prepare the lipid-modified concentrated acylglycerol,the content ofω-3 PUFAs in enzymatically concentrated tuna oil acylglycerol were significantly increased,while the oxidation stability was significantly decreased(P<0.05).
