中文摘要:以不同分子质量组分的马尾藻岩藻聚糖(SF)为研究对象,体外培养小鼠肉瘤细胞S-180,做抗肿瘤活性试验。采用MTT法测定不同铺板数量和不同浓度SF作用下的细胞生长曲线,确定细胞的铺板数量和SF对肿瘤细胞增殖抑制作用的有效浓度范围;测定不同组分SF在不同浓度、时间条件下对该细胞的增殖抑制率及对肿瘤细胞的IC50值。在倒置显微镜下观察SF2-R对细胞形态的影响。试验结果显示,当细胞铺板数量为5×103个/mL时,由细胞数量反映的OD值与时间的线性相关性良好;当SF的质量浓度设定在1~25 mg/mL范围时,细胞增殖抑制作用与SF浓度线性相关性良好;8种分子质量组分的SF对肿瘤细胞均有抑制作用,其中SF2-R对细胞的抑制作用最强。SF2-R对S-180肿瘤细胞的IC50为7.88 mg/mL,低于其他组分对肿瘤的IC50。倒置显微镜观察结果是:随着SF浓度的增加,细胞凋亡加重。

外文摘要:In this study, different fractions of fucoidan from Sargassum henslowianum were used as the object to do anti-tumor experiments in vitro by cultivating mouse sarcoma S180 cell. The effect of different cell counts in the planking and SF with different concentrations on cell growth curve was detected according to the MTT assay to determine cell counts in the planking and effective concentration range of SF about inhibitory effect of tumor cell proliferation. When SF with different components was under the condition of different concentrations and time, the inhibitory effect of this cell proliferation and ICs0 value on tumor cells were detected. The effect of SF2-R on morphology of cell was observed under an inverted microscope. The results show that, when cell count in the planking is 5×10^3/mL, the OD value reflecting from cell count and time have a good linear correlation. When concentration of SF is set in the range of 1-25 mg/mL, inhibitory effect of cell proliferation and concentration of SF have a good linear correlation. Eight kinds of SF with different components all have inhibitory effect on tumor cells, in which the inhibitory effect of SF2-R on cells is the strongest. IC50 value of SF2-R on S-180 tumor ceils is 7.88 mg/mL, which is lower than other components. It is found that state of cell apoptosis aggravates with the increase of the concentration of SF under an inverted microscope.
