中文摘要:【目的】研究丁香酚和MS-222对凡纳滨对虾(Litopenaeus vannamei)的麻醉效果,筛选对虾保活流通过程中适宜麻醉剂及其浓度、时间。【方法】分别配制100、150、200 mg/L的丁香酚和1、2、3 mL/L的MS-222两种麻醉液,将体质量(18.13±1.57)g、体长(8.25±1.73)cm的凡纳滨对虾放入其中,以入麻时间、呼吸频率、复苏时间及复苏率为评价指标,评价两种麻醉剂在11、14和18℃下对凡纳滨对虾的麻醉效果。【结果】水温18℃时,入麻时间随丁香酚和MS-222浓度升高而显著减少(P<0.05),复苏时间则呈剂量依赖性显著延长(P<0.05)。在15 min内,100、150、200 mg/L的丁香酚均可使呼吸频率由初始的(113.42±5.28)次/min降至(94.65±5.48)次/min,且复苏率100%,在100 mg/L的丁香酚和1 mL/L的MS-222两种麻醉液中,入麻和复苏时间随水温升高而显著缩短(P<0.05)。11℃下丁香酚适宜质量浓度为200 mg/L,麻醉时间不宜超过40 min;14℃下适宜质量浓度为150 mg/L,麻醉时间不宜超过30 min;18℃下适宜质量浓度为100 mg/L,麻醉时间不宜超过20 min。【结论】丁香酚较MS-222在不同实验浓度和温度条件下,均能快速降低凡纳滨对虾呼吸频率和行为应激反应,且复苏快、存活率高,是凡纳滨对虾麻醉的首选麻醉剂。
外文摘要:【Objective】To screen for the optimal concentration and exposure time of anesthetics on shrimp during live transport,the anesthetic efficacy of eugenol and MS-222 on Litopenaeus vannamei was comparatively studied.【Methods】Eugenol(100,150,200 mg/L)and MS-222(1,2,3 mL/L),were prepared with the normal seawater.Shrimp with the body weight of(18.13±1.57)g and length of(8.25±1.73)cm were immersed in the anesthetics,the temperature of 11,14 and 18℃.The induction and recovery time,respiration frequency,and recovery yields were determined and the anesthesia efficacy of eugenol and MS-222 was assessed accordingly.【Results】At 18℃,the shrimp were induced into anesthetic state in shorter than that at 11 and 14℃.Within 15 min,the respiratory frequency dropped from the initial value of(113.42±5.28)times per min to(94.65±5.48)times per min when induced by eugenol agent with concentration of 100,150,and 200 mg/L respectively,and the recovery rate was maintained at 100 percentage.In addition,the recovery time from anesthetic state was significantly prolonged with the increase of eugenol and MS-222 concentrations(P<0.05).When the shrimp was anesthetized in 100 mg/L of eugenol and 1 mL/L of MS-222 respectively,the time to anesthetic state or time for recovery was significantly reduced(P<0.05)as compared to those at higher concentrations of anesthesia.【Conclusion】In compassion with MS-222,shrimp immersed in eugenol exhibits higher survival rate and is proved to be a preferred anesthetic agent for L.vannmei.Moreover,eugenol concentration of 200 mg/L and induction time of 40 min was optimal at 11℃;the corresponding values were 150 mg/L and 30 min at 14℃;and the duration intensity of 100 mg/L and 20 min was appropriate at 18℃.
