中文摘要:试验旨在研究姜黄素对13月龄罗曼粉蛋鸡卵巢组织和Sirt1、Sirt2、Sirt3基因表达的影响。选取21只健康、体型相近的13月龄罗曼粉蛋鸡,其中1只鸡为对照组(CK组);剩余20只鸡随机分成4组(C1组、C2组、C3组、C4组),每组5个重复,每个重复1只鸡。各试验组饲喂基础日粮并分别灌喂低剂量(4 mg/kg,C1组)、中剂量(20 mg/kg,C2组)、高剂量(100 mg/kg,C3组)、超高剂量(500 mg/kg,C4组)的姜黄素油剂,对照组饲喂基础日粮和灌喂4 mL压榨葵花仁油。试验期为4周。结果显示:一定剂量的姜黄素能够改变蛋鸡卵巢组织形态结构,且卵泡内容物和着色点均有所增加,对产蛋后期蛋鸡的卵巢具有保护和修复组织形态的可能性。姜黄素能够有效提高卵巢中Sirt1、Sirt2、Sirt3的表达量;与CK组相比,C3组、C4组的Sirt1表达量显著提高(P<0.05);C1组、C2组的Sirt2基因表达量显著提高(P<0.05),C3组、C4组的Sirt2基因表达量极显著提高(P<0.01);C2组Sirt3基因表达量极显著提高(P<0.01),C1组、C3组的Sirt3基因表达量显著提高(P<0.05)。研究表明,姜黄素能够改变蛋鸡卵巢组织形态结构,并显著上调Sirt1、Sirt2、Sirt3基因的表达。

外文摘要:The purpose of the experiment was to study the effect of curcumin on ovarian tissue and Sirt1, Sirt2, Sirt3 gene expression in 13-month-old Roman pink laying hens. A total of 21 healthy and similar 13-month-old Roman pink laying hens were selected, one of which was control group(CK group), the remaining 20 chickens were randomly divided into four groups(C1 group, C2 group, C3 group, C4 group), with five replicates in each group and one hens in each replicate.Chickens in test group was fed with basal diet and fed with low dose(4 mg/kg, C1 group), medium dose(20 mg/kg, C2group), high dose(100 mg/kg, C3 group) and ultra high dose(500 mg/kg, C4 group) of curcumin oil, and control group was fed with basal diet and 4 mL of pressed sunflower oil. The trial period was 4 w. The results show that a certain concentration of curcumin can change the morphological structure of the ovary of laying hens, and the follicle content and coloring points are increased, which has the possibility of protecting and repairing the ovary of laying hens in the later stage of laying eggs. Curcumin can effectively increase the expression of Sirt1, Sirt2 and Sirt3 in ovary. Compared with CK group, the expression of Sirt1 in C3 group and C4 group was significantly increased(P<0.05). The expression of Sirt2 in C1 group and C2 group was significantly increased(P<0.05), and the expression of Sirt2 in C3 group and C4 group was extremely increased(P<0.01). The expression level of Sirt3 gene was extremely increased in C2 group(P<0.01), and the expression level of Sirt3 gene in C1 group and C3 group was significantly increased(P<0.05). The experiment indicates that curcumin can change the morphological structure of ovary in laying hens, and significantly up-regulate the expression of Sirt1, Sirt2 and Sirt3 genes.
