中文摘要:H9N2亚型禽流感病毒(AIV)是家禽和野鸟中流行的主要亚型之一,具有宿主范围广泛、持续流行的特点,不仅给家禽饲养业造成了严重的危害,而且给人类健康带来了日益严重的威胁。该亚型的病毒在AIV重组中扮演了供体基因的角色,在病毒的重组和跨宿主传播方面具有重要的生态学意义。为了解广东省雷州半岛家养水禽-鸭体内的H9N2亚型AIV的携带状况,本研究于2019年从雷州半岛东、西、南、北4个鸭场的568份样品中分离到3株H9N2亚型AIV,通过遗传演化分析发现,3株病毒均存在不同程度的重组现象。更重要的是,1株H9N2AIV GD/F2为鸡-鸭-野鸟-人的多重重组病毒。2株病毒GD/ML14和GD/F2的M基因来自于人源株A/Beijing/1/2017(H9N2)。这也是首次在家禽中发现如此复杂多样的重组病毒。3株病毒的受体结合位点均发生偏嗜向人源受体结合位点的突变,某些氨基酸的突变意味着对小鼠的致病性增强。总之,H9N2亚型AIV处于不断的变异之中,应该进一步加强针对H9N2亚型AIV的监测,为疫情防控提供数据支持。

外文摘要:The stable and persistent prevalence of H9N2subtype avian influenza virus(AIV)in poultry has not only caused serious harm to the poultry industry,but also posed a growing threat to human health.It has the characteristics of wide host range and persistent epidemic.This subtype of influenza virus could provide their inner genes to other subtypes,which played important ecological significance on reassortment and transmission among different species.In order to understand the H9N2subtype influenza virus in ducks of Leizhou peninsula in Guangdong Province,this study acquired three strains of H9N2subtype influenza virus from 568samples from 4duck farms in the east,west,south and north of Leizhou peninsula in 2019.Genetic evolution analysis revealed that all three strains showed different degrees of recombination.More importantly,a strain of H9N2influenza,GD/F2,was a five-fold reassortment virus of chicken-duck-wild bird-environmenthuman.The M genes of GD/ML14and GD/F2 were derived from human A/Beijing/1/2017(H9N2).This is the first time that such a complex and diverse recombinant virus has been found in poultry.All three strains have mutations in the receptor binding site that favor binding to the human receptor.In total,the H9N2subtype of AIV is in constant variation,so the surveillance of H9N2subtype should be further strengthened to provide data support for epidemic prevention and control.
