中文摘要:目的探讨珠母贝糖胺聚糖的抗肿瘤作用机制。方法采用鸡胚绒毛尿囊膜(CAM)模型观察珠母贝糖胺聚糖单用及与5-氟尿嘧啶(5-FU)合用对CAM血管生成的影响。结果珠母贝糖胺聚糖CPG、PGⅠ、PGⅡ在实验所选剂量(0·5、5、50、500mg·L-1)下,无明显的直接抑制CAM血管生成作用;但CPG、PGⅠ、PGⅡ在实验所选剂量(5、50mg·L-1)下与5-FU(2·5mg·L-1)合用时,均能增敏5-FU的抑制CAM血管生成作用(P<0·01),并呈一定的剂量效应关系,其中PGⅡ增敏效果较好。结论珠母贝糖胺聚糖通过抑制肿瘤血管生成而对5-FU的抗肿瘤作用起增敏效果。
外文摘要:Aim To study the antitumor mechanism of glycosaminoglycan from Pinctada martensii. Methods In vivo, the effects of glycosaminoglycan from Pinctada martensii on neovascularization was observed by chick chorioallantoic membrane (CAM) model. Re' suits CPG. PG Ⅰ . PG Ⅱ had no obvious anti-angiogenesis ability at the dosage of 0. 5 - 500 mg· L^-1, but they could enhance the anti-angiogenesis ability of 5-FU (2. 5 mg· L^-1 ) at the dosage of 5 - 50 mg · L^-1( P 〈 0. 01 ) in a concentration-dependent manner. The anti-angiogenesis ability of PG 11 was better than that of CPG and PG I. Conclusions The glycosaminoglycan from Pinctada martensii has the effect of antiangiogenesis, which may be one mechanism of the synergistic effect of 5-FU on cancer cells.
