中文摘要:为了揭示中国荷斯坦奶牛低密度脂蛋白受体相关蛋白5(low density lipoprotein receptor-related protein 5,LRP5)基因单核苷酸基多态性(SNP)与中国荷斯坦奶牛乳品质性状之间的关联,试验提取98头中国荷斯坦奶牛DNA,采用直接测序技术来检测LRP5基因多态性位点,对其SNP位点不同基因型与乳品质性状进行关联分析。结果显示,在LRP5基因第4内含子上发现3个SNPs位点,分别是I4-90C>T、I4-229C>A和I4-570A>G。其中I4-90C>T存在TT、CC和TC基因型,TC基因型尿氮素显著高于TT和CC基因型(P<0.05)。然而,该位点的突变与产奶量和脂肪含量无显著相关性(P>0.05);I4-229C>A存在CC和CA 2个基因型;I4-570A>G存在AA、GG和GA基因型。2个突变位点与中国荷斯坦奶牛乳品质性状无显著相关性(P>0.05)。结果表明,LRP5基因I4-90 C>T位点可作为影响中国荷斯坦奶牛乳品质性状的分子标记应用于奶牛的标记辅助选择。
外文摘要:In order to reveal the association between single nucleotide-based polymorphism(low density lipoprotein receptor-related protein 5,LRP5),DNA of 98Chinese Holstein cows were extracted and the LRP 5gene polymorphism locus was detected by direct sequencing,and related different genotypes and milk quality traits at the SNP loci was analyzed.The results showed that three SNPs were found on the fourth intron of the LRP 5gene,I4-90C>T,I4-229C>A and I4-570A>G.Among them,I4-90C>T had TT,CC and TC genotypes,and TC genotype was significantly higher than TT and CC genotypes(P<0.05).However,mutations at this locus were not significantly associated with milk yield and fat content(P>0.05);genotypes CC and C for I4-229 C>A;genotypes AA,GG and GA for I4-570A>G.The two mutation sites showed no significant correlation with milk quality traits in Chinese Holstein dairy cows(P>0.05).The above results indicate that the I4-90C>T site of LRP 5gene can be used for marker-assisted selection as molecular markers affecting milk quality traits in Chinese Holstein cows.
