中文摘要:以中巨石斑鱼精巢组织为原料,优化提取工艺条件,提取鱼精蛋白,测定抗菌活性蛋白的分子量,并对其抗菌活性进行评价。主要结果如下:(1)通过正交优化试验,对鱼精蛋白的提取工艺进行了优化,最佳提取工艺条件是硫酸浓度为10%、硫酸用量为精巢质量的4倍、提取时间为1.5 h、冷乙醇用量为硫酸提取液的3.5倍,在此条件下,鱼精蛋白得率达1.24%,抗菌活性达到54.99%;(2)通过Tricine-SDS-PAGE电泳对抗菌活性蛋白的分子量进行测定,主体成分蛋白的分子量小于16 ku,在(13~14)ku之间;(3)鱼精蛋白对枯草芽孢杆菌和大肠杆菌均具有较好的抑菌活性,其对枯草芽孢杆菌的抑菌活性比大肠杆菌更显著。

外文摘要:In this study, the testis tissue of Giant grouper was used as a raw material to extract its antimicrobial protein. The extraction technology was optimized. The molecular weight and antibacterial activity of the protein were determined. The main results are the followed:(1) The extraction process of antimicrobial protein from Giant grouper's testis were optimized by an orthogonal test. The results showed that the optimal conditions were sulfuric acid of 10% concentration, the volume of sulfuric acid 4 times of raw materials mass, 1.5 h of extraction time, and the volume of cold ethanol 3.5 times the volume of sulfuric acid solution. Under this condition, the extracted rate of the protein was the highest, which reached up to 1.24%. The antimicrobial activity reached up to 54.99%.(2) The molecular weight of the main antimicrobial protein measured by Tricine-SDS-PAGE electrophoresis is below 16 ku, which located between 13 ku and 14 ku.(3) The extracted protein showed strong antibacterial activity against Bacillus subtilis and Escherichia coli, the antibacterial protein showed a better antibacterial activity against Bacillus subtilis.
