中文摘要:【目的】分析北部湾两个野生皮氏叫姑鱼的群体遗传多样性和遗传结构,为其种质资源的合理开发利用提供参考依据。【方法】利用已开发的皮氏叫姑鱼微卫星标记对北部湾临高县西海域(N20°20',E109°40)'和乐东县西海域(N19°30',E107°80)'的两个野生群体进行遗传多样性和遗传结构分析。【结果】筛选出的7个微卫星位点中有4个位点(Jobe50、Jobe45、Jobe24、Jobe13)呈多态性。4个多态位点的等位基因数(No)介于10~13个,平均10.75个,期望杂合度(He)为0.772~0.844,观测杂合度(Ho)为0.827~0.892,多态信息含量(PIC)为0.824~0.847。两个皮氏叫姑鱼群体各多态位点的群体内近交系数(Fis)为-0.114~0.015,总群体近交系数(Fit)为-0.049~0.031,群体内有明显的杂合过剩现象;群体遗传分化系数(Fst)为0.016~0.082,基因流(Nm)为2.804~28.236。【结论】两个北部湾野生皮氏叫姑鱼群体均具有中等遗传多样性,有明显杂合过剩的现象,群体间存在较大的基因流,群体遗传分化水平较低,临东县西海域群体的遗传多样性水平高于乐东县西海域群体。
外文摘要:【Objective】The population diversity and genetic structure of two wild Beibu Gulf Johnius belengerii varieties were studied in order to provide theoretical reference for the reasonable exploitation and utilization of relevant germplasm resources. 【Method】Microsatellite DNA markers were used to analyze the genetic diversity and structure of 2 wild populations of Johnius belengerii captured from the West Sea of Lingao County (N 20°20', E 109°40') and West Sea of Ledong County (N 19°30', E 107°80'). 【Result】Four of the seven microsatellite locus (Jobe50, Jobe45, Jobe24, Jobe13) were polymorphic. The number of alleles from the 4 polymorphic locus (No) varied from 10 to 13 with the average at 10.75, the expected heterozygosities (He) varied from 0.772 to 0.844, the observed heterozygosities (Ho) varied from 0.827 to 0.892, and the polymorphism information content (PIC) varied from 0.824 to 0.847. In the two wild Johnius belengerii varieties, the inbreeding population polymorphism coefficient (Fis) was between -0.114-0.015, the total inbreeding coefficient (Fit) was between -0.049-0.031, so the groups were obviously observed with the phenomenon of heterozygous excess; the genetic differentiation coefficient (Fst) was between 0.016-0.082, and gene flow (Nm) was between 2.804-28.236. 【Conclusion】There were mediocre genetic diversity and excess of heterozygote in the two Johnius belengerii populations. Between the populations, there existed large genetic flow and low levels of genetic differentiation, in which the genetic diversity of the population from the West Sea of Lindong County was higher than that of the population from the West Sea of Ledong County.
