中文摘要:本研究旨在探讨不同槲皮素含量对凡纳滨对虾(Litopenaeus vannamei)抗副溶血弧菌(Vibrio parahaemolyticus,VP)性能的影响。选取凡纳滨对虾[(3.65±0.93)g],每100克商品饲料中分别添加0(对照组)、0.1、1.0、2.0 g槲皮素,进行为期14 d的饲喂试验,并在饲喂结束后测定肝胰腺及肠道的抗氧化酶活性、免疫相关酶活性以及免疫相关基因的表达水平;随后进行副溶血弧菌攻毒试验,评估不同槲皮素含量对凡纳滨对虾抗VP性能的影响。结果表明:1.0 g槲皮素组凡纳滨对虾的存活率显著高于0.1、2.0 g槲皮素组(P<0.05)。随着饲料中槲皮素含量的升高,凡纳滨对虾肝胰腺中超氧化物歧化酶(SOD)活性显著升高(P<0.05),其中1.0、2.0 g槲皮素组SOD活性显著高于对照组、0.1 g槲皮素组(P<0.05),但1.0、2.0 g槲皮素组间SOD活性无显著差异(P>0.05)。1.0 g槲皮素组凡纳滨对虾肝胰腺组织溶菌酶(LZM)、谷胱甘肽过氧化物酶(GSH-Px)和碱性磷酸酶(AKP)活性显著高于对照组和0.1、2.0 g槲皮素组(P<0.05)。VP攻毒后,随着饲料中槲皮素含量的升高,凡纳滨对虾肝胰腺LZM活性显著升高(P<0.05),其中1.0、2.0 g槲皮素组显著高于对照组、0.1 g槲皮素组(P<0.05),但1.0、2.0 g槲皮素组间无显著差异(P>0.05)。1.0 g槲皮素组SOD、GSH-Px和AKP活性显著高于0.1、2.0 g槲皮素组(P<0.05)。攻毒后,1.0 g槲皮素组凡纳滨对虾肝胰腺和肠组织免疫相关基因Toll样受体(Toll-like Receptor)、抗脂多糖因子(Anti-lipopolysaccharide Factor)、溶菌酶(Lysozyme)、组织蛋白酶B(Cathepsin B)、甲壳素(Crustin)和转录因子激活蛋白-1(Transcription Factor AP-1-like)基因的相对表达量显著高于0.1、2.0 g槲皮素组(P<0.05)。综上所述,每100克饲料中添加1.0 g槲皮素能显著增强凡纳滨对虾抗VP性能。槲皮素有望用作凡纳滨对虾的功能性饲料添加剂,可降低因VP感染所导致的死亡率。

外文摘要:This study investigated the effects of different quercetin levels in diets on the resistance of Litopenaeus vannamei to Vibrio parahaemolyticus(VP).The L.vannamei individuals weighing(3.65±0.93)g were fed the diets supplemented with 0(control),0.1,1.0,and 2.0 g quercetin per 100 g feed for 14 d.After feeding,the antioxidant enzyme activities,immune-related enzyme activities,and immune gene expression in the hepatopancreas and intestine were measured.Subsequently,L.vannamei individuals were challenged with VP to evaluate the resistance.The results showed that L.vannamei fed 1.0 g quercetin had significantly higher survival rate than those fed 0.1 or 2.0 g(P<0.05).The superoxide dismutase(SOD)activity in the hepatopancreas increased as the quercetin level increased(P<0.05),being significantly higher in the 1.0 g and 2.0 g groups than in the 0 and 0.1 g groups(P<0.05),with no difference between the 1.0 g and 2.0 g groups(P>0.05).The activities of lysozyme(LZM),glutathione peroxidase(GSH-Px),and alkaline phosphatase(AKP)in the hepatopancreas in the 1.0 g group were significantly higher than those in the 0.1 g and 2.0 g groups(P<0.05).After VP challenge,the LZM activity in the hepatopancreas enhanced with the increase in quercetin level(P<0.05),being significantly higher in the 1.0 g and 2.0 g groups than in the 0 and 0.1 g groups(P<0.05),with no significant difference between 1.0 g and 2.0 g groups(P>0.05).The SOD,GSH-Px,and AKP activities in 1.0 g group were significantly higher than those in the 0.1 g and 2.0 g groups(P<0.05).Moreover,the relative expression of immune genes encoding Toll-like receptor,anti-lipopolysaccharide factor,LZM,cathepsin B,crustin,and transcription factor AP-1-like in the hepatopancreas and intestine in the 1.0 g group was significantly higher than that in the 0.1 g and 2.0 g groups(P<0.05).These results indicate that dietary supplementation with 1.0 g quercetin per 100 g feed effectively enhances VP resistance in L.vannamei.Quercetin has the potential as a functional feed additive to reduce VP-induced mortality in L.vannamei.
