中文摘要:确定黑桑葚脂质体的最佳制备工艺,研究其表征与评价,探究桑葚花青素脂质体应用于开发天然植物绿色染发剂产品的实现。提取桑葚花青素,采用逆向蒸发法和pH梯度法制备花青素脂质体,采用单因素实验考察大豆卵磷脂与胆固醇质量比(为1∶4、1∶5、1∶6、1∶7、1∶8)、溶解壁内膜有机溶剂的种类(乙醚、石油醚、三氯甲烷、二氯甲烷、乙醇、乙酸乙酯、乙醚∶乙醇=1∶1)、桑葚花青素冻干粉加入量(0.5、1.0、1.5、2.0、2.5mg/mL)对包封率的影响;为优化以得到最佳的桑葚花青素脂质体制备工艺条件,设计桑葚花青素冻干粉加入量(1.0、1.5、2.0 mg/L)、胆固醇与卵磷脂的质量比值(1∶5、1∶6、1∶7)及溶解壁内膜脂有机溶剂的种类(二氯甲烷、三氯甲烷、乙醚)三因素三水平实验,确定花青素脂质体的最佳制备工艺,对该工艺制得的脂质体进行表征。得到桑葚花青素脂质体最佳制备工艺条件为以乙醚为溶解膜脂的有机溶剂、桑葚花青素冻干粉的加入量为1.5mg/mL(即18 mg)、胆固醇与卵磷脂的质量比值为1∶5。在最佳制备工艺条件下进行验证性实验,得到所制备花青素脂质体包封率达72.23%。
外文摘要:To determine the best preparation process of black mulberry liposome, study its characterization and evaluation, and explore the realization of application of mulberry anthocyanin liposome in the development of natural plant green hair dye products. Extract mulberry anthocyanins, prepared by reverse evaporation method and pH gradient anthocyanins liposomes, adopt single factor experiment investigation quality of soybean lecithin and cholesterol than(1, 1∶5,1∶6, 1∶7, 1∶8), dissolving wall lining the kinds of organic solvents(ether, petroleum ether, chloroform, dichloromethane, ethanol, ethyl acetate, ethyl ether,Ethanol =1∶1), mulberries anthocyanin freeze-dried powder dosage(0.5, 1.0, 1.5, 2.0, 2.5 mg/mL) on the encapsulation rate;In order to optimize and obtain the optimal preparation conditions of mulanthocyanin liposomes, a three-factor and three-level experiment was designed, including the addition amount of mulanthocyanin freeze-dried powder(1.0, 1.5, 2.0 mg/L), the mass ratio of cholesterol to lecithin(1∶5, 1∶6, 1∶7) and the types of organic solvents(dichloromethane, trichloromethane, ether) to dissolve the inner wall lipid. The optimum preparation process of anthocyanin liposome was determined and the liposome prepared by this process was characterized. The optimum preparation conditions were obtained as follows: ether was used as the organic solvent to dissolve membrane lipid, the dosage of mulanthocyanin freeze-dried powder was 1.5 mg/mL(18 mg), and the mass ratio of cholesterol to lecithin was 1∶5. The encapsulation efficiency of the prepared anthocyanin liposome was 72.23 %.
