中文摘要:试验旨在研究白花鬼针草水提物、甲醇提取物和乙醇提取物的化学成分及其抗氧化性和抗菌性。以水、80%甲醇和50%乙醇分别作为提取剂获得提取物,通过黄酮类、蒽醌类、萜类等成分的颜色反应、沉淀反应等化学预试法分析提取物成分并研究其总黄酮类物质提取效率差异;采用DPPH法、ABTS法和羟基自由基清除能力测定法来评价3种提取物的体外抗氧化性能;测定3种提取物对金黄色葡萄球菌、蜡状芽孢杆菌、沙门氏菌和大肠杆菌的最小抑菌浓度(MIC)及最小杀菌浓度(MBC)。结果表明:白花鬼针草含有黄酮类、强心苷、酚类、鞣质类等多种活性物质,主要为黄酮类化合物,其中水提取物中含有的活性成分更丰富;以50%乙醇作为提取剂时白花鬼针草中总黄酮类物质提取效果最佳,水次之,80%甲醇则最差,提取率分别为13.34mg/g、8.10mg/g和2.44mg/g;50%乙醇提取物具有良好的抗氧化性,其对羟基自由基、DPPH和ABTS自由基的清除率分别达94.41%、85.94%和99.84%;对同一病原菌而言,抑菌活性最好的是甲醇提取物,最弱的是水提取物;对同一提取物而言,沙门氏菌最敏感,而金黄色葡萄球菌则耐受性最强。综合来看,白花鬼针草50%乙醇提取物具有良好的抗氧化及抗菌效果,可为开发利用白花鬼针草资源提供参考。
外文摘要:The aim of this experiment was to study the chemical constituents,inoxidizability and antibacterial activity of water extract,methanol extract and ethanol extract from Bidens alba.Water,80%methanol and 50%ethanol were used as extracting agents to obtain extracts respectively.Compositions of these extracts were detected by chemical pretest including color reaction and precipitation reaction of flavonoids,anthraquinones,terpenoids and other chemical constituents,and extraction efficiency of the total flavonoids was analyzed.The methods of DPPH,ABTS and hydroxyl radical scavenging capacity were adopted to evaluate the antioxidant properties of the three extracts in vitro.The minimal inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of the three extracts against Staphylococcus aureus,Bacillus cereus,Salmonella and Escherichia coli were further determined.The results show that the plant contains many kinds of active substances,such as flavonoids,cardiac glycosides,phenols and tannins,mainly flavonoids,among which the water extract contains more active components.When 50%ethanol was used as the extractant,the extraction effect of total flavonoids from Bidens alba was the highest,followed by water,and 80%methanol was the lowest.The extraction rates were 13.34 mg/g,8.10 mg/g and 2.44 mg/g,respectively.The scavenging rate of 50%ethanol extract was 94.41%for hydroxyl radical,85.94%for DPPH radical and 99.84%for ABTS radical.For the same pathogen,methanol extract had the best antibacterial activity,while water extract had the weakest antibacterial activity.For the same extract,Salmonella was the most sensitive bacteria,while Staphylococcus aureus was most resistant to extract.It can be concluded that the 50%ethanol extract of Bidens alba possesses nice antioxidant and antibacterial effects,which can provide reference for the development and utilization of Bidens alba.
