中文摘要:为了探讨依托泊苷诱导的2.4蛋白(etopositde-induced 2.4protein,EI24)基因在雷州黑鸭群体中的遗传多态性,分析与雷州黑鸭产蛋性状相关的潜在遗传标记位点,本试验以雷州黑鸭为研究对象,采用PCR扩增技术获得EI24基因的目的片段,通过直接测序法检测到突变位点,随后使用GraphPad Prism 6.0软件对产蛋性状和基因型与单倍型之间进行多重比较,分析EI24基因的SNP位点与雷州黑鸭产蛋性能的相关性。结果表明,EI24基因内含子5中发现7个SNP位点,其中,I5-17G>T、I5-59G>A>T、I5-159G>A、I5-110T>A和I5-256C>A位点与雷州黑鸭开产日龄显著相关;I5-166G>A和I5-385C>T位点与雷州黑鸭开产体质量极显著相关。单倍型分析发现,H1H4单倍型可作为雷州黑鸭群体筛选低开产日龄、高开产体质量、高产蛋量个体的综合最优单倍型。说明EI24基因多态性可为选育优势高产蛋量的雷州黑鸭提供有利的遗传标记。

外文摘要:The aim of this study is to investigate the genetic polymorphism of the etopositde-induced 2.4protein(EI24)gene in Leizhou black duck population,and to analyze the potential genetic marker loci related to egg-laying traits of Leizhou black duck.Taking Leizhou black duck as the research object,the target fragment of the EI24gene was obtained by PCR amplification technology,and the mutation site was detected by direct sequencing method.GraphPad Prism 6.0software was used to conduct post hoc multiple comparisons between egg laying traits,genotypes and haplotypes and SPSS 22.0software were used to analyze the correlation between the SNP locus of the EI24gene and the egg-laying performance of Leizhou black ducks.The results showed that 7SNP sites were found in intron 5of EI24gene,among which,I5-17G>T,I5-59G>A>T,I5-159G>A,I5-110T>A and I5-256C>A loci were significantly correlated with the Leizhou black duck birthing age;the I5-166G>A and I5-385C>T loci were associated with the birth weight of the Leizhou black duck very significantly correlated.The results showed that the H1H4haplotype could be used as the comprehensive optimal haplotype for the selection of individuals with low start-to-lay age,high start-to-lay weight and high egg production in the Leizhou black duck population,indicating that the EI24gene polymorphism can provide favorable genetic markers for breeding Leizhou black ducks with high egg production.
