外文摘要:The nutrients, chitin and pigments resources in black soldier fly (Hermetia illucens) (BSF) recently attracts attention. However, current procedure for sequential extraction of these BSF resources in industrial scale remains not clearly established. Our study developed an extraction process for peptide, chitin and melanin based on defatted BSF (DFBSF). The results showed that enzymatic hydrolysis with alkaline protease and keratinase could reduce the DFBSF substrate by 81.05 % (dry basis). After hydrolysis, the mixture was filtered and the hydrolysate was collected and dried to yield BSF peptide (BSFP), and then melanin and chitin were extracted from the remained insect residue. The melanin extraction process was optimized as liquid-to-solid ratio of 15:1, NaOH 2.2 M, and temperature of 70 degrees C for 90 min to obtain the maximum yield. Chitin was then extracted from the solid fraction after the melanin extraction. The BSFP was characterized by 61.86 % crude protein, 54.09 % total amino acids, and 88.30 % of peptides with molecular weights (Mw) below 1000 Da. The spectral, elemental composition and HPLC properties indicated that BSF melanin mainly consists of eumelanin. The absorption bands of IR spectra at 1650, 1619, and 1551 cm- 1 indicated that BSF chitin belongs to alpha-crystalline chitin. A 50 kg scaling-up trial was conducted to validate the industrial feasibility of this process. Through this process, 50.00 kg of DFBSF was transformed into 32.24 kg BSFP, 0.85 kg melanin and 5.77 kg chitin. The establishment of BSFP, chitin and melanin extraction provided the insect farming industry with efficient conversion of biomass into high valueadded materials.
