中文摘要:为了鉴定杂交种子与父母本的亲缘关系,用20条随机引物和21条ISSR引物在供试的一组三系杂交水稻F1、父母本及对照进行扩增。20条RAPD引物共扩增出78个条带,其中28条为多态性条带;21条ISSR引物共扩增出72个条带,其中多态性条带为20条。将这些多态性条带进行聚类分析,F1与父、母亲缘关系较近归为一类,随后它们才与对照归为一类。结果表明RAPD和ISSR引物的PCR扩增均能有效地鉴定三系杂交水稻的杂种F1与组合中父、母本的遗传关系。与RAPD相比ISSR引物在三系杂交水稻及其父母本的鉴定中具有稳定性更好的优点。
外文摘要:The DNA of three-line hybrid rice F1, its parents and the check were amplified by 20 random primers and 21 ISSR primers in order to identify the relationship of F1 and it parents. 78 bands,in which 28 bands were polymorphic, were amplified with 20 random primers, 72 bands,in which 20 bands were polymorphic,were amplified with 21 ISSR primers. The polymorphic bands were hierarchical cluster analyzed, F1 was first grouped into one with its parents lines,then it was grouped into one with the check. The results showed that the relationship of F1 and parents' line could be identified effectively by amplification of random primers and ISSR primers. The resuits of ISSR were stabler than those of random primers in identification of three-line hybrid rice and its parents.
