中文摘要:为了研究中国荷斯坦奶牛丝氨酸蛋白酶抑制剂E2(serpin peptidase inhibitor, clade E, member 2,SERPINE2)基因的多态性及与乳品质性状的关系,试验采用PCR扩增及DNA测序的方法检测了128头中国荷斯坦奶牛SERPINE2基因的多态性,分析了单核苷酸多态性(SNPs)位点的连锁不平衡情况,探讨了各基因型、单倍型与乳品质性状的关系。结果表明:中国荷斯坦奶牛SERPINE2基因的外显子2和内含子2上共存在4个SNPs位点,分别为E2-203 G>A、I2-166 A>G、I2-170 T>C、I2-377 G>A,这4个SNPs位点的基因杂合度(H)和多态信息含量(PIC)均在0.25~0.50之间,处于哈代温伯格平衡状态(P>0.05)。E2-203 G>A位点AA基因型个体305 d校正产奶量显著高于GG、GA基因型(P<0.05);I2-166 A>G位点AA基因型个体乳糖含量显著高于GA基因型(P<0.05),GG基因型个体305 d校正产奶量显著高于GA基因型(P<0.05);I2-170 T>C位点TT、CC基因型个体乳糖含量显著高CT基因型(P<0.05),CC基因型个体305 d校正产奶量显著高于CT基因型(P<0.05);I2-377 G>A位点GG基因型个体乳糖含量显著高于GA、AA基因型(P<0.05),AA基因型305 d校正产奶量显著高于GA基因型(P<0.05)。H3H3单倍型组合个体的305 d产奶量显著高于H1H1、H1H2、H2H2单倍型组合(P<0.05)。说明E2-203 G>A、I2-166 A>G、I2-170 T>C、I2-377 G>A位点可作为中国荷斯坦奶牛乳品质性状的相关分子标记,H3H3单倍型个体的产奶量高于其余单倍型,可作为奶量性状的最优单倍型。
外文摘要:In order to study the polymorphism of serpine protease inhibitor E2(SERPINE2) gene and its relationship with milk quality traits, PCR amplification and DNA sequencing technology were used to study the polymorphism of SERPINE2 gene in 128 Chinese Holstein dairy cows, and the linkage imbalance was analyzed to explore the correlation between the haplotype, genotype and milk quality traits. The results showed that there were four SNPs locus in exon 2 and intron 2 of SERPINE2 gene in Chinese Holstein dairy cows, which were E2-203 G>A, I2-166 A>G, I2-170 T>C, and I2-377 G>A, respectively. The heterozygosity(H) and polymorphic information content(PIC) of the four SNPs loci ranged from 0.25 to 0.50, which were in the Hardy-Weinberg equilibrium state(P>0.05). The AA genotypes individuals of E2-203 G>A locus were significantly higher than GG in corrected milk quantity on day 305, GA genotype(P<0.05), and the AA genotypes individuals of I2-166 A>G locus were significantly higher than AG genotype(P<0.05) in the lactose content, GG genotype individuals were significantly higher than AG genotype(P<0.05) in corrected milk quantity on day 305;TT and CC genotype individuals of I2-170 T>C locus were significantly higher than CT genotype(P<0.05) in lactose content, CC genotype individuals were significantly higher than CT genotype(P<0.05) in corrected milk quantity on day 305;GG genotype individuals of I2-377 G>A locus were significantly higher than that of GA, and AA genotype(P<0.05) in the lactose content, AA genotype individuals was significantly higher than GA genotype(P<0.05) in corrected milk quantity on day 305. The milk yield of H3 H3 haplotype combination individuals on day 305 was significantly higher than that of H1 H1, H1 H2 and H2 H2 haplotype combination individuals(P<0.05). The results indicated that the SNP sites of E2-203 G>A, I2-166 A>G, I2-170 T>C, I2-377 G>A could be used as molecular markers related to milk quality traits in Chinese Holstein dairy cows. The milk yield of H3 H3 haplotype was higher than that of other haplotypes, which could be used as the optimal haplotype for milk yield traits.
