中文摘要:以金花茶浓缩液为试验材料,通过乙醇沉淀、透析等方法分离得到金花茶粗多糖TPS。采用CelluloseDE-52离子交换柱层析对金花茶粗多糖进行分离纯化,得到TPS0、TPS1、TPS2、TPS3、TPS4和TPS56个糖组分,并对各糖组分的理化性质进行初步分析。糖分析结果显示,金花茶粗多糖的半乳糖醛酸约为中性糖的一半,表明金花茶粗多糖中含有相当一部分果胶物质;TPS0、TPS1和TPS23个组分以中性糖为主,而TPS3、TPS4和TPS53个糖组分则含有较高比例的半乳糖醛酸,表明TPS0、TPS1和TPS23个糖组分为中性多糖,TPS3、TPS4和TPS53个糖组分为果胶物质。构成糖分析结果显示,金花茶粗多糖中主要的构成糖有葡萄糖、半乳糖、阿拉伯糖和甘露糖,它们以不同比例分布于离子交换柱层析并分离纯化得到的各糖组分;TPS3、TPS4和TPS53个糖组分中均含有约11%的鼠李糖,说明这3个果胶组分很可能由以半乳糖醛酸与鼠李糖交替连接而成的果胶分子链的毛发区域和由n个半乳糖醛酸连接而成分子链的平滑区域组成。另外,以上3个糖组分中均含有10%左右的葡萄糖,这与文献报道的果胶物质中葡萄糖比例较低有较大差异。
外文摘要:A crude polysaccharide matter (TPS) was obtained from leaves of Camellia chrysantha (Hu) Tuyama by alcohol precipitation and dialysis steps. Then the TPS was isolated and purified by a Cellulose DE-52 column chromatography, and six fractions(TPS0, TPS1, TPS2, TPS3, TPS4 and TPS5) were obtained. Sugar analysis showed that the content of neutral sugar was two times of galacturonic acid, indicated that TPS contained some pectins with high ratio of neutral sugar. Neutral sugars were the major sugar components in TPS0, TPS1 and TPS2 fractions, while the ratios of galacturonic acid in TPS3, TPS4 and TPS5 fractions were obviously high. This result indicated that TPS0, TPS1 and TPS2 fractions were almost some neutral polysaccharides, while TPS3, TPS4 and TPS5 fractions were some pectic substances. Sugar composition analysis showed that glucose, galactose, arabinose, and mannose were the major compositions in TPS, which distributed in fractions from TPS for various ratios. There were approximately 11% rhamnose in TPS3, TPS4 and TPS5 fractions, indecated that these 3 fractions containing a hair area considered as a bond chain linked by galacturonic acids and rhamnoses for a repeating type and a smooth area linked by galacturonic acids. In the other hand, there were also 10% gluctose in the 3 fractions, this result was remarkable different from relative literatures.
