中文摘要:旨在探讨组氨酸解氨酶(histidine ammonia-lyase,HAL)基因在雷州黑鸭群体中的遗传多态性,分析雷州黑鸭产蛋性状相关的潜在遗传标记位点。以雷州黑鸭为研究对象,采用PCR扩增技术获得HAL基因的目的片段,通过直接测序法检测到突变位点,最后使用SPSS 22.0软件分析HAL基因的遗传多态性位点与雷州黑鸭产蛋性能的相关性。结果显示,HAL基因内含子1和内含子2中发现7个SNP位点,其中,I1-288 C>T、I1-373 T>G和I2-168 A>G位点与雷州黑鸭300 d产蛋量显著相关,使用HaploView软件对SNPs位点进行连锁分析发现H1H1单倍型的300 d产蛋数显著高于H1H3单倍型。结果表明,H1H1单倍型可作为雷州黑鸭群体筛选低高产蛋量个体的最优单倍型。本研究可为选育出高产蛋量的雷州黑鸭提供有利的分子遗传标记,为雷州黑鸭在分子标记辅助育种方面提供了理论依据。

外文摘要:This experiment investigated the genetic polymorphism of the histidine aminamonia-lyase(HAL)gene in Leizhen black duck.The potential genetic labeling sites related to egg laying of Leizhen black duck were analyzed.PCR were used to obtain the HAL gene.The mutant position was detected by direct sequencing,and SPSS 22.0 software was used to analyze the correlation between the genetic polymorphism of HAL gene and egg production performance.The results showed that:seven SNP loci were found in intron 1 and intron 2 of the HAL gene,among them,I1-288 C>T,I1-373 T>G,and I2-168 A>G loci were significantly correlated with 300 d eggs production of the Leizhou black ducks.Using HaploView software to carry out linkage analysis on SNP loci,it was found that the number of eggs production in 300 d of H1 H1 haplotype was significantly higher than that of H1 H3 haplotype.In summary,the H1 H1 haplotype can be used as the optimal haplotype for the Leizhou black duck population to screen individuals with low and high egg production.This study provides promising molecular genetic markers for breeding Leizhou black ducks with high egg production and an important theoretical basis for molecular marker-assisted breeding of Leizhou black ducks.
