中文摘要:乌贼墨多糖(Squid ink polysaccharides,SIP)是近年来发现的一种新型海洋生物活性多糖,在动物模型实验中发现具有抗氧化及化疗保护作用。本文主要通过乌贼墨作为原料,经酶法提取及Sephdex G-100色谱分离纯化得到一种含3%硫酸根的多糖,经高效液相及光谱分析显示,多糖纯度均一,分子量约为37ku。以SIP为细胞保护剂,环磷酰胺(Cyclophosphamide,CP)为化疗药物,昆明小白鼠为模型动物来分析SIP对动物化疗损伤的保护效果。结果表明:SIP能缓解CP导致小鼠体重(p<0.01)、睾丸重量(p<0.01)、睾丸指数(p<0.05)的降低,显著提高LDH活力(p<0.05)及降低γ-GT活力(p<0.01)。结论:SIP对CP导致睾丸的损伤保护效果显著。

外文摘要:Squid ink polysaccharides (SIP), a newly found marine bioactive glycosaminoglycon has been recently proved to have antioxidant ability and chemotherapy-protective activity on model animals.In this paper, a type of SIP containing 3% sulfate group was harvested from squid ink with protease digestion and Sephadex G-100,the molecular weight was about 37ku based on HPLC. Male Kunming mice exposed to chemotherapeutic drug cyclophosphamide(CP) were administered with SIP by gavage, which was used to assess the interventional effects of SIP as a cytoprotector on chemotherapy associated damage in murine tesficular tissue. The results showed that SIP alleviated the decrease of mice weight ( p 〈 0.01 ), testis weight ( p 〈 0.01 ) and testis index (p 〈0.05)caused by CP, and markedly elevated the activity of LDH (p 〈 0.05)and reduced the activity of γ-GT (p〈O.O1)in testes of CP-treated mice, which indicated significant chemotherapy-protective effects of SIP on murine testis.
