中文摘要:本研究从湛江流沙港马氏珠母贝养殖群体中挑选不同壳色个体为亲本,共构建了黑(BS)、红(RS)、黄(YS)和白壳色(WS)4个壳色选系F1,然后分别在4个壳色选育系F1中随机取样,利用3对AFLP引物分析其遗传结构与遗传分化。结果表明,每对引物的扩增位点数在104~109之间,共得到331个扩增位点;BS、RS、YS和WS壳色选系F1的多态位点比例分别为49.2%、49.5%、51.6%和63.4%,Shannon's多样性指数分别为0.1884、0.1886、0.1896和0.1954,4个壳色选育系F1的遗传分化系数(Fst)为0.1972。本研究说明经过一代壳色纯化4个壳色选育系F1出现显著遗传分化,也为马氏珠母贝壳色系选育或选育系间杂交提供了参考依据。

外文摘要:In this research, we constructed black, red, yellow and white four shell color selective lines of F1 by selecting different shell color individual as parents from reared populations of pearl oyster Pinctada martensii in the Liusha stock of Zhanjiang, which were designated as BS, RS, YS and WS lines, respectively. Then we randomly sampled F1 individual from four shell color selective lines respectively to analyze the genetic structure and variation by using AFLP molecular marker with three primers. The results showed that the number of amplification loci in each polymorphism primer was from 104 to 109 and we totally obtained 331 reproducible bands. The polymorphic locus percentages of the BS, RS, YS and WS lines were 49.2%, 49.5%, 51.6% and 63.4%, respectively and the Shannon's genetic diversity values ofBS, RS, YS and WS lines were 0.188 4, 0.188 6, 0.189 6 and 0.195 4, respectively, the genetic differentiation among these four shell color selective lines was 0.197 2. The present results illuminated that these four shell color selective lines displayed remarkably genetic variation after one-generation selection for shell color, moreover, the results would also provide a refference for selecting shell color line or crossing between the selective lines of pearl oyster P. mctrtensii.
