中文摘要:以金花茶花为研究对象,通过对其提取物及分离组分的活性成分进行分析比较,结合小鼠耳肿胀实验优选最佳抗炎的活性组分。该实验利用超声辅助提取工艺制备金花茶花提取物(Camellia nitidissima Chi flower extract,CNFE),并经XAD1600大孔树脂分离洗脱,依次得到水洗组分(CNFE.A)、30%(体积分数)乙醇组分(CNFE.B)、70%乙醇组分(CNFE.C)和95%乙醇组分(CNFE.D),并测定上述5个组分中总黄酮、总糖、总酚和总皂苷的含量。同时以雄性昆明小鼠为研究对象,通过二甲苯致小鼠耳肿胀模型来考察上述5个组分的抗炎作用。结果表明,CNFE中活性成分主要是黄酮类化合物和糖类化合物,含量分别为(37.20±4.70)g/100 g和(34.97±4.30)g/100 g。在二甲苯致小鼠耳肿胀实验中,CNFE对二甲苯致小鼠耳肿胀有显著抑制作用(P<0.05),抑制率为34.01%,CNFE.B对小鼠耳肿胀有极显著的抑制作用(P<0.01),抑制率为61.79%,并能够减少炎症细胞的浸润和显著降低血清中IL-6、IL-1β、TNF-α的分泌,其抗炎效果还优于吲哚美辛。金花茶花提取物中主要是黄酮类化合物,具有一定抗炎作用,30%乙醇洗脱组分为最佳抗炎组分。
外文摘要:Taking Camellia nitidissima Chi flower as the research object,the best anti-inflammatory active ingredients were selected by analyzing and comparing the active ingredients of the extracts and isolated components combined with mouse ear swelling experiments.Camellia nitidissima Chi flower extract(CNFE)were prepared by ultrasonic-assisted extraction and separated by XAD1600 macroporous resin column to obtain water elution fraction(CNFE.A),30%ethanol elution fraction(CNFE.B),70%ethanol elution fraction(CNFE.C),and 95%ethanol elution fraction(CNFE.D),and the contents of total flavonoids,total sugars,total phenols,and total saponins in the above five components were determined.At the same time,the anti-inflammatory effects of the above five fractions were also investigated in male Kunming mice through xylene-induced ear swelling in mice.Results showed that the active ingredients in CNFE were mainly flavonoids and glycoconjugates,the contents were(37.20±4.70)g/100 g and(34.97±4.30)g/100 g,respectively.In the xylene-induced mice ear swelling test,CNFE had a significant inhibitory effect on xylene-induced mice ear swelling(P<0.05),and the inhibition rate was 34.01%.CNFE.B had a highly significant inhibitory effect on ear swelling in mice(P<0.01),with an inhibition rate of 61.79%,and was able to reduce the infiltration of inflammatory cells and significantly decrease the secretion of IL-6,IL-1β,and TNF-αin serum,and its anti-inflammatory effect was also superior to that of indomethacin.Camellia nitidissima Chi flower extract are mainly flavonoids with certain anti-inflammatory effects,and 30%ethanol eluted fraction is the best anti-inflammatory fraction.
