中文摘要:【目的】从海洋环境中分离并筛选具有抑菌效果的细菌,为后续海洋源细菌素挖掘提供重要的微生物资源基础。【方法】收集湛江滨海区域的海水、海底泥和新鲜海藻样品。分别使用M10培养基、P3培养基、BHI培养基分离样品中的细菌,并利用固体琼脂打孔法从分离细菌中筛选对大肠埃希菌(Escherichia coli)、铜绿假单胞菌(Pseudomonas aeruginosa)、金黄色葡萄球菌(Staphylococcus aureus)和单增李斯特氏菌(Listeria monocytogenes)有抑菌活性的菌株。对具有抑菌活性的菌株进行16S rRNA基因的PCR扩增,并与近缘物种进行系统发育分析。【结果】分别从海水、海底泥和新鲜海藻中分离得到496、421和649株细菌,其中有抑菌活性的菌株数分别为49、48和60。共筛选出27株具有广谱抑菌活性的菌株,来源于海水的有5株(J-19-2、S-2-2、S-3-2、S-6-6和S-14-1),来源于海底泥的有6株(A05-2、A17-2、A20-6、B15-3、C10-4和C25-3),以及来源于新鲜海藻的有16株(A4-4、A4-5、A4-6、A4-11、A6-5、A7-2、A20-5、A21-4、A21-7、A25-6、B7-1、B25-1、C7-19、C9-11、C15-2和D2-5)。16S rRNA系统发育分析结果表明,16株具有广谱抑菌效果的海洋细菌属芽孢杆菌属(Bacillus),占比59.26%。此外,海水、海底泥和新鲜海藻样品中细菌群落的组成存在显著差异,但芽孢杆菌属在具有抑菌活性的细菌中均为优势菌属,所占比例分别为16.33%、71.92%和63.33%。【结论】湛江海域海洋环境拥有丰富的具有抗菌活性的微生物资源,其中芽孢杆菌属细菌抑菌潜能最突出。
外文摘要:【Objective】To isolate and screen marine bacteria with antibacterial activity from the marine environment,providing important microbial resources for the subsequent exploration of marine-derived bacteriocins.【Methods】Seawater,marine sediment,and fresh seaweed samples were collected from the coastal area of Zhanjiang.Bacteria were isolated using M10 medium,P3 medium,and BHI medium,respectively,and the antibacterial activity of the isolates against Escherichia coli,Pseudomonas aeruginosa,Staphylococcus aureus,and Listeria monocytogenes were determined by the agar well diffusion method.The 16S rRNA gene of strains with antibacterial activity were amplified by PCR,and their phylogeny with closely related species were analyzed.【Results】A total of 496,421,and 649 bacterial strains were isolated from seawater,marine sediment,and fresh seaweed,respectively,of which 49,48,and 60 strains exhibited antibacterial activity.27 strains with broad-spectrum antibacterial activity were screened,including J-19-2,S-2-2,S-3-2,S-6-6,and S-14-1 from seawater;A05-2,A17-2,A20-6,B15-3,C10-4,and C25-3 from marine sediment;and A4-4,A4-5,A4-6,A4-11,A6-5,A7-2,A20-5,A21-4,A21-7,A25-6,B7-1,B25-1,C7-19,C9-11,C15-2,and D2-5 from fresh seaweed.The 16S rRNA phylogenetic analysis showed that 59.26%(16/27)of the marine bacteria with broad-spectrum antibacterial activity belonged to the genus Bacillus.In addition,there were significant differences in bacterial community composition among seawater,marine sediment,and seaweed samples,but Bacillus was the dominant genus among antibacterial strains,accounting for 16.33%,71.92%,and 63.33%,respectively.【Conclusion】The marine environment of the Zhanjiang coastal area possesses abundant microbial resources with antibacterial activity,among which Bacillus genus show the most prominent antibacterial potential.
