中文摘要:非特异性细胞毒性细胞(non-specific cytotoxic cell,NCC)是硬骨鱼中一类淋巴细胞,在非特异性免疫中通过上调炎症反应释放细胞因子发挥重要的作用,是哺乳动物中NK细胞的等价物。鱼类NCC在体内多个组织器官中均有分布,其细胞毒性是自发的,可杀死多种靶细胞,例如同种异源或异种异源肿瘤细胞,寄生性原生动物和被病原侵染的细胞。NCCRP-1是NCC上的一种标志性受体。本研究对尼罗罗非鱼的脑,头肾,肝脏和脾脏进行切片,对NCCRP-1进行染色,通过免疫组化分析得知NCC在这些组织中的分布情况。另外采用Percoll非连续密度梯度法分离纯化得到头肾NCC,采用免疫组化染色和FITC标签标记,与组织中的染色结果分布相同。相较于qPCR等技术,本研究更直观地观察NCC在组织中的分布特点,为深入研究NCC的功能奠定了基础。

外文摘要:It is well known that n on-specific cytotoxic cell is a kind of lymphocyte in teleosts, which plays an important role in nonspecific immune by up-regulating inflammation responses to release cytokines. NCCs are equivalents of NK cells in mammals. They distribute in tissues and organs with spontaneous cytotoxicity which can kill various target cells such as allogeneic or xenogeneic tumor cells, parasitic protozoan and cells induced by pathogens. NCCRP-1 is a landmark receptor on NCCs. We sectioned brain, head kidney, liver and spleen of Nile Tilapia(Oreochromis niloticus) and stained NCCRP-1 as well. The study obtained the distribution of NCCs in tissues via immunohistochemical analysis. Furthermore, we also isolated and obtained NCCs of head kidney through Percoll density gradient centrifugation, stained by immunohistochemistry and tagged with FITC. The distribution was the same as the staining result in tissues. This study observed the distribution characteristics of NCCs in tissues more clearly than q PCR technique, which might lay the foundation for further studies on the function of NCC.
