中文摘要:于我国雷州半岛红树林海区捕获隶属于弹涂鱼属(Periophthalmus)、大弹涂鱼属(Boleophthalmus)与青弹涂鱼属(Scartelaos)的4种弹涂鱼,克隆4种弹涂鱼23个个体cox1基因序列,通过分析4种弹涂鱼与Gen Bank中12种弹涂鱼cox1基因序列的变异特征,探讨4种弹涂鱼与后者的种间亲缘关系。结果表明,16种弹涂鱼的82个体的cox1基因序列共检测到单倍型44个,核苷酸多态位点212个(占35.4%),种间平均核苷酸差异数为4(占2.01%);基于最大似然法(ML)和邻接法(NJ)构建的系统发育树显示,各弹涂鱼属多形成一独立分支,亲缘关系明显,齿弹涂鱼属(Periophthalmodon)镶嵌于弹涂鱼属内,大弹涂鱼属与青弹涂鱼属形成并列分支,亲缘关系较近。依化石记录标定的松散分子钟估算物种分歧时间,结果表明,各弹涂鱼属出现明显分化大约发生于渐新世末期至中新世早期(23.61~15.65 Ma)。

外文摘要:Four species of mudskippers, including Periophthalmus, Boleophthalmus and Scartelaos,were caught in the mangrove area of China’s Leizhou Peninsula, and 23 cox1 gene sequences of these mudskippers were cloned. By analyzing the variation characteristics of mitochondrial cox1 genes and phylogenetic relationship of 16 species from the Leizhou Peninsula, China and GenBank in other seaside, the efficacy of 82 mitochondrial cox1 genes was evaluated as DNA barcodes for mudskippers.The research showed that genetic diversity in 82 homologous cox1 genes of 599 bp was evaluated with 44 haplotypes, 212 nucleotide polymorphism loci (35.4%) and 4 average number of nucleotide differences (2.01%). Combing the genetic distances of taxa with the phylogenetic relation by using the neighbor joining (NJ) and maximum likelihood (ML), this article illustrated most of Genus formed a monophyletic group, such as Periophthalmodon embedded in Periophthalmus, Boleophthalmus and Scartelaos formed a paraphyletic group. The relaxed molecular clock was calibrated by fossil records to estimate the divergence date. These estimates suggested that those Genus, including Periophthalmus,Periophthalmodon and others, separated about 23.61 – 15.65 Ma ago in the late Oligocene to the Early Miocene.
