中文摘要:目的:研究波纹巴非蛤糖胺聚糖体外清除自由基作用及对衰老模型小鼠的体内抗氧化作用。方法:采用体外实验研究波纹巴非蛤糖胺聚糖对羟自由基、超氧阴离子自由基、DPPH·的清除作用。体内实验采用小鼠腹腔注射D-半乳糖建立衰老模型,同时给药组灌服高、中、低3个剂量的波纹巴非蛤糖胺聚糖溶液,4周后检测各组小鼠超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)含量。结果:波纹巴非蛤糖胺聚糖精制品(PUG-1)体外对羟自由基、超氧阴离子自由基、DPPH自由基均具有较好的清除作用,IC50分别为:4.23、3.19、4.36mg/mL。与衰老模型组小鼠比较,200、400mg/kg·dPUG-1可以极显著提高衰老小鼠血清、肝脏、脑组织中SOD、GSH-Px和CAT含量(p<0.01),降低MDA含量(p<0.05)。结论:波纹巴非蛤糖胺聚糖具有体外清除自由基作用,其体内抗衰老作用可能与提高机体抗氧化酶活性有关。
外文摘要:Objective.The free radical scavenging capacities and the antioxidant effect in vivo of glycosaminoglycan from Paphia undulate( PUG and PUG-1 )in aging mice were researched.Methods:The scavenging effect to the hydroxyl radical,superoxide radical and DPPH ~ were detected with the oxidation of 1,10-phenanthroline-Fe2+, self-oxidation of pyrogallic acid system and DPPH · system, respectively. Aging model mice caused by 200mg/kg· d D-galactose were.irregated with 100,200 and 400mg/kg · d PUG-1 for 4 weeks,the effect of PUG-1 on the content of superoxide dismutase ( SOD ), Glutathione peroxidase ( GSH- Px), malondialdehyde ( MDA), catalase(CAT) in serum,liver and brain were observed.Results:PUG-1 had strong scavenging effect on hydroxyl radical,superoxide radical,and DPPH ~ radical with IC5o of 4.23,3.19 and 4.36mg/mL, respectively. In vivo,the activities of SOD,GSH-Px and CAT in serum, liver and brain in aging model mice were lower and the content of MDA that was higher compared to the normal mice( p 〈0.01).While in the aging model mice treated with PUG-1 200 and 400mg/kg ~ d,the activity of SOD,GSH-Px and CAT were significantly increased and the content of MDA decreased( p 〈 0.05 or p 〈 0.01 ).Conclusion. PUG-1 had free radical scavenging effect.The anti-aging function maybe related to the enhancement of antioxidase activity.
