中文摘要:为探究热应激环境对蛋鸡血清外泌体miRNA表达谱的影响。选取40周龄健康状况良好,生产性能相近的罗曼粉壳蛋鸡40只,随机分为两组,常温对照组(NC组)保持鸡舍处于(25±3)℃,高温控制组(HC组)环境温度控制在(32±3)℃。在严格控制环境温度、湿度及光照时间下,饲养30 d后,进行血清的分离、外泌体的提取及鉴定,测定血清外泌体浓度变化,利用深度测序技术分析蛋鸡血清外泌体miRNA差异表达谱,并利用实时荧光定量PCR(qRT-PCR)技术分析了部分差异表达的miRNA。结果显示,高温控制组蛋鸡血清外泌体浓度较常温对照组极显著提高(P<0.01),测序数据进行分析显示,高温控制组相比常温对照组有83个miRNA表达差异,对其中9个差异miRNA定量验证结果与测序数据一致;对两组表达差异的83个miRNA靶基因功能进行分析,主要涉及的调控通路有氧化还原反应、半胱氨酸和蛋氨酸代谢、氧化磷酸化、脂肪酸降解、细胞内蛋白质运输、蛋白质转运、细胞凋亡。试验结果为研究热应激对蛋鸡血清外泌体miRNA表达谱的影响、发现新的热应激诊断生物标志物、提高蛋鸡生产性能提供了参考数据。
外文摘要:To explore the effect of heat stress on serum miRNA expression profile of laying hens.Forty 40-week old Lohmann laying hens with good health and similar production performance were randomly divided into two groups,the normal temperature control group(NC group)kept the henhouse at(25±3)℃,and the high temperature control group(HC group)kept the ambient temperature at(32±3)℃.Under strict control of ambient environmental temperature,humidity and light duration.The serum samples were used to extract and identify exosomes,and then detected the concentration of serum exosomes.The differential expression profile of serum exosomes miRNA was analyzed by deep sequencing technology.Some of the differentially expressed miRNAs were verified by quantitative real-time fluorescence PCR(qRT-PCR).The results showed that the concentration of exosomes in serum of HC group was higher than NC group(P<0.01).According to the sequencing data,83 miRNAs expression differences were found in the HC group compared with NC group.Nine of these miRNAs expression analyzed by qRT-PCR were consistent with the sequencing data.The function of these miRNAs target genes was analyzed,and the regulatory pathways closely related to redox process,cysteine and methionine metabolism,oxidative phosphorylation,fatty acid degradation,intracellular protein transport,protein export and apoptosis.The results are usefull for study the effect of heat stress on the expression profile of serum exosomes in laying hens and find new biomarkers for diagnosis of heat stress.It provides important reference data for improving production performance of laying hens.
