中文摘要:目的探究虾头酶解物对灌服辣椒素小鼠炎性因子、肠道菌群及脂多糖的影响。方法选用30只C57BL/6J雄性小鼠,随机分为空白对照组、辣椒素组、鱼油阳性对照组以及辣椒素+虾头酶解物低、中、高[0.45、0.90、1.80 g/(kg·d)]组,每组5只。灌服7 d后,以ELISA法测定血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)和白细胞介素-10(interleukin-10,IL-10)含量,以16S rRNA基因高通量测序法测定小鼠粪便中肠道菌群的结构组成,以鲎试剂法测定血清中细菌脂多糖(lipopolysaccharide,LPS)含量。结果高剂量辣椒素可以使血清中的TNF-α和IL-1β等炎症标志物水平增加2倍以上,成功构建系统性低度炎症模型。辣椒素致炎模型小鼠服用低、中、高剂量虾头酶解物和实验剂量鱼油后,血清中炎症标志物TNF-α分别下调52%、54%、47%和33%,接近空白对照组水平。另外,低、高剂量虾头酶解物可以增加辣椒素致炎模型小鼠的肠道菌群多样性,降低变形菌门/厚壁菌门的比例。在属水平上,降低嗜麦芽窄食单胞菌属和棒状杆菌属等有害菌丰富度,增加阿洛巴菌属和乳杆菌属等有益菌的丰富度。同时,显著降低革兰阴性菌和潜在致病菌的丰富度及血清中LPS水平。结论虾头酶解物可以通过调节肠道菌群结构,降低血清中LPS水平,缓解由辣椒素诱导的小鼠系统低度炎症,效果优于鱼油。

外文摘要:Objective To investigate the effect of shrimp head enzymatic hydrolysate(SHEH)on systemic low-grade inflammation of mice induced by capsaicin.Methods A total of 30 C57 BL/6 J male mice were randomly divided into 6 groups,including blank control group,capsaicin group,fish oil positive control group,and capsaicin+SHEH low dose,medium dose and high dose[0.45,0.90,1.80 g/(kg·d)]groups,respectively.After 7 days of gavage,the serum levels of inflammatory markers TNF-α,IL-1β,IL-6 and IL-10 were measured with ELISA,the concentration of lipopolysaccharide(LPS)was determined with the microplate quantitative chromogenic matrix limulus kit,and gut microbiota in the fecal samples were measured with 16 S rRNA gene high-throughput sequencing.Results High-dose capsaicin increased the levels of inflammatory markers such as TNF-αand IL-1βin the serum by more than two times,indicating a successful construction of systemic low-grade inflammation models.After capsaicin-induced inflammation model mice were given low,medium and high doses of SHEH and experimental dose of fish oil,the serum inflammation marker TNF-αwas respectively reduced by 52%,54%,47%and 33%respectively,which was close to the level in blank control group.In addition,low and high doses of SHEH increased the diversity of gut flora of capsaicin-induced inflammation model mice.The ratio of Proteobacteria/Firmicutes decreased significantly.At the genus level,the abundances of Stenotrophomonas and Corynebacterium generally decreased,while those of Allobaculum and Lactobacillus increased significantly.At the same time,the abundances of Gram-negative bacteria and pathogenic bacteria in the gut of mice and the concentration of LPS in the serum significantly decreased.Conclusion Shrimp head enzymatic hydrolysate can decrease the concentration of LPS in the serum by adjusting the structure of gut microbiota,alleviating the systemic low-grade inflammation of mice induced by capsaicin,with a better effect than fish oil.
