中文摘要:为了研究酰基辅酶A硫脂酶12(acyl-CoA thioeaterase 12,ACOT12)基因的多态性及其与肉品质性状的关系,本试验采用DNA测序技术对102头四川牦牛ACOT12基因的多态性进行了研究,使用Haploview 4.2软件对ACOT12基因的多态性位点进行连锁不平衡分析,并通过SPSS 23.0软件分析ACOT12基因的SNPs位点及其单倍型与肉品质性状的相关性。结果发现,四川牦牛ACOT12基因外显子1上存在3个SNPs位点,分别为E1-285 T>C、E1-603 T>A、E1-675 C>G,其中E1-603 T>A、E1-675 C>G位点分别与pH_(45 min)值、背膘厚性状显著相关(P<0.05),说明以上2个SNPs可分别作为四川牦牛宰后pH_(45 min)和背膘厚性状的相关分子标记;H2H2单倍型组合的宰后pH_(45 min)值显著高于其余H1H4单倍型组合,因此可作为四川牦牛宰后pH_(45 min)值性状的最优单倍型组合,为未来优质肉牛的早期选育提供分子理论依据。
外文摘要:In order to study the polymorphism of acyl-CoA thioeaterase12(ACOT12)gene and its relationship with meat quality traits,DNA sequencing technique was used to study the polymorphism of ACOT12gene of Sichuan yak,Haploview 4.2software was used to analyze the linkage disequilibrium of ACOT12gene polymorphism,and SPSS 23.0software was used to analyze the correlation between SNPs locus and haplotype of ACOT12gene and meat quality traits.The results showed that there were three SNPs loci on exon 1of ACOT12gene in Sichuan yak,which were E1-285T>C,E1-603T>A and E1-675C>G,respectively.Among them,E1-603T>A and E1-675 C>G were significantly correlated with pH_(45min)and backfat thickness traits,respectively,indicating that the above two SNPs could be used as molecular markers for postmortem pH_(45min)and backfat thickness traits in Sichuan yak.The postmortem pH_(45min)value of H2H2haplotype combination was significantly higher than that of other H1H4haplotype combinations,so it could be used as the best haplotype combination for postmortem pH_(45min)value traits of Sichuan yak,and provide molecular theoretical basis for early breeding of high-quality beef cattle in the future.
