中文摘要:【目的】探究马氏珠母贝(Pinctada fucata martensii)对干露胁迫的响应,为马氏珠母贝健康养殖和遗传育种提供基础资料。【方法】在不同温度下对马氏珠母贝进行干露胁迫,探究温度对马氏珠母贝干露耐受的影响;筛选出马氏珠母贝干露胁迫敏感组和耐受组,比较两组的免疫相关基因表达量和植核后休养期存活率。【结果】(1)22~34℃条件下,马氏珠母贝干露耐受能力随温度升高及干露时间延长而降低;(2)22℃下,S组和T组的脯氨酸羟化酶(PHD)、葡萄糖转运蛋白(GLUT)、髓样分化因子(MYD88)、热休克蛋白(Hsp70、Hsp20)和凋亡抑制因子(cIAP)6个基因在干露过程中表达量基本呈上升趋势,组间差异显著(P<0.05);(3)植核试验结果表明耐受组植核后休养期的存活率显著高于敏感组(P<0.05)。【结论】马氏珠母贝在高温环境下干露耐受能力降低,在适温条件下耐受组比敏感组表现出较高的存活率。

外文摘要:【Objective】To analyze the response of Pinctada fucata martensii to air exposure stress and to understand its environmental adaptability.【Method】Air exposure of Pinctada fucata martensii was carried out at different temperatures to explore the influence of temperature on the tolerance of air exposure.At 22℃,the immune-related gene expression levels and the resting survival rate of the two groups were compared in the stress-sensitive and stress-tolerant groups.【Result】(1)Under 22-34℃,the air exposure toleranceof P.f.martensii decreased with increasing temperature;(2)Under 22℃,the expression level of proline hydroxylase(PHD),glucose transporter(GLUT),myeloid differentiation factor(MYD88),heat shock protein(Hsp70,Hsp20)and inhibitor of apoptosis(cIAP)6 genes in the S and T groups showed an upward trend basically,and the difference between the groups was significant(P<0.05);(3)The survival rate of the stress-tolerant group during the recuperation period after nucleation was significantly higher than that of the stress-sensitive group(P<0.05).【Conclusion】The tolerance to air exposure was reduced under high temperature environment,and the stress-tolerance group showed higher survival rate than the stress-sensitive group under suitable temperature conditions.
