中文摘要:为了探索香泽兰总黄酮对斑马鱼脾脏显微组织结构的毒性作用,采集香泽兰全草,用70%乙醇(v/v)浸泡后,辅以超声提取,再经大孔树脂柱层析纯化,浓缩,冰冻干燥,制得香泽兰总黄酮冻干粉,用此总黄酮冻干粉按不同浓度对斑马鱼进行水触媒染毒,21 d后取脾脏,制作石蜡切片,苏木精-伊红染色,显微观察。结果发现,随着香泽兰总黄酮水触媒染毒浓度的增加,供试斑马鱼脾脏显微组织结构出现了不同程度的病变,主要表现含铁血黄素沉着增多,脾窦扩张,脾脏内淋巴细胞增多,血细胞核浓缩、溶解,甚至导致脾脏坏死。说明一定浓度的香泽兰总黄酮对斑马鱼脾脏具有一定的毒性作用。

外文摘要:To probe into the micro-structural changes of spleen of zebrafish exposure to flavonoids extracted from Eupatorium odoratum Linn.. the whole natural E.odoratum was collected, dried and ground, then its flavonoids were extracted by 70% ethanol (v/v) marinating and ultrasonic technology. Afterwards, the extract was purified by macroporous resin and made into frozen-dried powder. Different concentrations cff the frozen-dried powder of flavonoids were exposed to zebrafish (Danio rerio) by water pollution. After 21 days of exposure, the spleen of the zebrafish was collected to make paraffin-clothed and hematnxylin-eosin stained slides, and the histopathological changes were observed. It was found that, with the dosage increasing of the flavonoids, the microstructure of spleen of the zebrafish exposed to the flavonoids appeared pathological changes as follows: increase of hemosiderosis, dilated splenic sinus, increase of lymphocytes, condensed and dissolved nuclear of blood cell, and even necrosis of spleen. It was concluded that certain concentrations of flavonoids extracted from E.odoratum caused certain toxicities to spleen of zehrafish.
