中文摘要:目的:研究环境条件对腐败希瓦氏菌生物被膜形成的影响。方法:采用改良的96微孔板法,通过改变环境因素,研究不同培养时间、菌体浓度和理化因子对腐败希瓦氏菌生物被膜形成能力的影响。用玻璃试管法和扫描电镜观察,以不锈钢片或聚乙烯塑料(PE)片为接触面,对腐败希瓦氏菌形成生物被膜能力及形态结构的影响。结果:腐败希瓦氏菌在LB培养基中静置培养约24 h,形成成熟的生物被膜。在各实验条件下,以30℃培养,2%NaCl,pH中性或略偏碱性,起始菌浓度106~107CFU/mL时形成生物被膜的量最大;而Ca2+和一定浓度的Mg2+能抑制腐败希瓦氏菌生物被膜的形成。葡萄糖的添加量对生物被膜的形成没有明显的影响。腐败希瓦氏菌在不锈钢片上形成的生物被膜的量高于在PE片上形成的量,其形态分别为网络状、矩阵态。结论:腐败希瓦氏菌能形成稳定而明显的生物被膜,且其形成能力受温度、NaCl浓度、pH值等环境因子的影响。接触面影响对腐败希瓦氏菌生物被膜形成的能力及形态结构。本研究结果为通过食品加工环境对希瓦氏菌生物被膜的有效控制提供理论支持。

外文摘要:Objective: Shewanella putrefaciens as one of the specific spoilage organisms (SSO) of Litopenaeus van- namei, which was chosen in this study to research the characters of biofilm. Method: Microtiter-plate method was used to stimulate biofilm formation under different environmental conditions. What's more, glass test tube method and scanning electron microscopy were used to research the ability and pattern of biofilms formatted on stainless steel sheet and polyethylene plastic sheet (PE), respectively. Results: The results shows that Shewanella putrefciens formed integral biofilms at 30 ℃ under 2% NaCl concentration, the concentration of bacterial suspension of 106-107 CFU/mL, the neutral and alkalescence condition, culturing for 24 h in LB. Addition of Ca^2+ and a certain concentration of Mg^2+ obviously inhibited the biofilm formation while glucose had no apparent effect on it. The evidence that biofilm on stainless steel sheet was much more than it on PE was provided by glass test tube method. And it could be watched that the biofilms are like network-like and matrix state which were formatted on the stainless steel sheet and PE respectively through scanning electron microscopy. Conclusion: It can be summarized that a stable and number of biofilms on contact surfaces which display different patterns can be formatted by Shewanella putrefaciens, and biofilm formation is affected by environmental factors such as temperature, the concentration of NaCl, the value of pH and so on. Furthermore this paper provides the- oretical guidance to control ShewaneUa putrefaciens biofilm effectively.
