中文摘要:旨在优化双水相萃取法提取马友鱼肝中乙酰胆碱酯酶的工艺条件,并为乙酰胆碱酯酶的提取提供理论依据。双水相系统由三种离散盐和两种分子量的聚乙二醇(PEG)两两组合优化,最终选取(NH_(4))_(2)SO_(4)/PEG1000双水相体系提取马友鱼肝中的乙酰胆碱酯酶(AChE),并通过正交试验得到最佳萃取条件。最佳实验条件:PEG-1000和(NH_(4))_(2)SO_(4)的质量分数分别为19%和25%,pH值为7.0,温度为20℃。在此条件下,乙酰胆碱酯酶的回收率为77.25%。双水相萃取法简化了大马哈鱼肝脏中乙酰胆碱酯酶的提取过程,且工艺条件经过优化后实现了提取率的有效提高。

外文摘要:This paper is aimed to optimize the aqueous two-phase extraction process for the extraction of acetyl cholinesterase(AChE)from the liver of eleutheronema tetradactylum,and provide a theoretical basis for the AChE extraction.There were three discrete salts and two molecular weights polyethylene glycol(PEG),the aqueous two-phase system was optimized by the combination between one discrete salt and one molecular weights PEG,the (NH_(4))_(2)SO_(4)/PEG-1000 aqueous two-phase system was selected to extract the AChE from the liver of Eleutheronema tetradactylum,and the extraction conditions were optimized by orthogonal experiment.The optimal conditions were as follows:mass fractions of PEG-1000 and (NH_(4))_(2)SO_(4) were 19%and 25%,pH 7.0,extraction temperature 20℃.Under these conditions,the recovery rate of AChE was 77.25%.The aqueous two-phase extraction method simplifies the extraction process of AChE from the liver of king salmon and increases the extraction rate effectively.
