中文摘要:基于生物秀丽隐杆线虫模型探究卡拉胶寡糖的抗衰老能力。采用含不同质量浓度(0.25、0.50、1.00 mg/mL)卡拉胶寡糖的大肠埃希氏菌液(OP50)饲喂线虫,以饲喂OP50菌液组作为空白对照,观察卡拉胶寡糖对线虫的寿命、繁殖力、体长体宽、运动能力和应激能力等健康指数的影响,以及测定线虫体内活性氧(reactive oxygen species,ROS)含量和抗氧化酶活性,探究卡拉胶寡糖对线虫抗衰老的影响。与对照组线虫相比,各浓度的分子质量为1.58 kDa的卡拉胶寡糖(KCO-5)均可延长线虫寿命,能将线虫的寿命延长4.18%~16.48%;改善了线虫的体长体宽、运动和应激能力,降低了其体内脂褐素积累水平,且不会损伤繁殖能力。KCO-5能够显著降低线虫体内ROS和丙二醛含量,并提高了总超氧化物歧化酶、过氧化氢酶和还原型谷胱甘肽的活性,且呈剂量依赖性(P<0.05)。结果表明,KCO-5中和了过量产生的ROS,增强了线虫的抗逆性,并延长了寿命。综上,卡拉胶寡糖具有抗衰老功效,是减少氧化应激、改善健康寿命的功能食品或药物替代品的一种新的可能来源。
外文摘要:This study aimed to explore the anti-aging ability of carrageenan oligosaccharides based on the biobeauty Caenorhabditis elegans model.Nematodes were fed with Escherichia coli solution containing different concentrations(0,0.25,0.50,1.00 mg/mL)of carrageenan oligosaccharides.The study observed the effects of carrageenan oligosaccharides on nematode longevity,fecundity,body length,body width,lipofuscin,exercise capacity,and heat stress capacity and measured the reactive oxygen species(ROS)content and antioxidant enzyme activity in nematodes,investigating the effect of carrageenan oligosaccharides on anti-aging in nematodes.Compared with the control group,all concentrations of carrageenan oligosaccharide(KCO-5)with a molecular weight of 1.58 kDa could prolong the lifespan of nematodes and could extend the lifespan of nematodes by 4.18%-16.48%.It improved the body length,width,locomotion and stress capacity of nematodes,reduced the level of lipofuscin accumulation in the body,and did not impair reproductive ability.KCO-5 significantly reduced the content of ROS and malondialdehyde in nematodes and increased the activities of superoxide dismutase,catalase,and glutathione in a dose-dependent manner(P<0.05).Results showed that KCO-5 neutralized the excess ROS,enhanced the stress resistance of nematodes,and extended the lifespan.In summary,carrageenan oligosaccharides have anti-aging benefits and are a new possible source of functional food or drug alternatives for reducing oxidative stress and improving a healthy lifespan.
