中文摘要:【目的】分析马氏珠母贝酶解蛋白粉(Enzymolysis protein powder of Pinctada martensii,EPP)营养组成,并探讨EPP改善自然衰老斑马鱼学习记忆能力的功效及其可能的作用机制。【方法】采用国标法测定EPP营养组成。18月龄斑马鱼分模型组和实验组,另取6月龄斑马鱼作对照组,实验组饲喂EPP[3 g/(kg?d)]30 d,其它组喂食正常饲料,通过T迷宫试验观察各组斑马鱼学习记忆能力,采用酶联免疫吸附法(ELISA)检测鱼脑组织中炎症因子IL-1β、TNF-α、IL-10含量及脑源性神经营养因子(Brain derived neurotrophic factor,BDNF)水平。【结果】EPP含有质量分数78.28%的蛋白质,富含谷氨酸、天冬氨酸、赖氨酸、亮氨酸、精氨酸等氨基酸,其钙、铁、锌元素含量较高。与模型组相比,EPP可以显著提高实验组斑马鱼觅食成功率及右臂游动路程占比(P<0.05),降低潜伏期;与模型组相比,EPP可以降低实验组斑马鱼脑组织炎症因子IL-1β、TNF-α含量(P<0.05),分别降低了3.18、2.77 pg/mg,同时提高其BDNF的水平(P<0.05),提高了54.99 pg/mg,但对IL-10含量无显著影响(P>0.05)。【结论】EPP具有改善自然衰老斑马鱼学习记忆能力的功效,其机制与EPP缓解大脑的炎症状况、提高脑源性神经营养因子水平有关。

外文摘要:【Objectives】To elucidate the nutritional composition of enzymolysis protein powder of Pinctada martensii(EPP),and investigate the improving effect of EPP on the learning and memory ability of natural aging zebrafish and its possible mechanism.【Methods】The nutritional composition of EPP was measured by the national standard method.18-months-old zebrafish were randomly divided into 2 groups:model group and experimental group,and 6-month-old zebrafish were taken as the control group.The treated group was fed EPP at a dose of 3 g/(kg?d)for 30 days,and the other groups were fed normal feed.The T maze experiment was employed to observe the learning and memory abilities of zebrafish.The levels of inflammatory factors IL-1β,TNF-α,IL-10 and brain-derived neurotrophic factor(BDNF)in the brain were determined by the enzyme-linked immunosorbent assay(ELISA).【Results】EPP contains 78.28%of protein,which is rich in glutamic acid,aspartic acid,lysine,leucine and arginine.It also has high contents of calcium,iron and zinc.Compared with the model group,EPP significantly increased the ethological success rate and the proportion of right arm travel distance of aging zebrafish(P<0.05),reduced the latency period.Compared with the model group,EPP reduced the levels of inflammatory factors IL-1βand TNF-αin the brain of aging zebrafish(P<0.05)by 3.18 and 2.77 pg/mg,respectively.Also,it can increase the level of BDNF by 54.99 pg/mg(P<0.05),while it had no significant effect on the content of IL-10(P>0.05).【Conclusion】EPP can improve the learning and memory ability of aging zebrafish.The mechanism is that EPP ameliorates the inflammation of the brain and increase the level of brain-derived neurotrophic factors.
