中文摘要:牛脂多糖结合蛋白(lipopolysaccharide binding protein,LBP)基因是脂质转移蛋白家族成员之一,其与细胞表面受体结合,启动先天宿主反应,同时参与机体脂质代谢。为了揭示LBP基因多态性与中国荷斯坦奶牛乳脂性状的相关性,本试验以133头中国荷斯坦奶牛为研究对象,采用基因测序法对待测奶牛第二、五内含子目的片段进行多态性研究,使用SPSS 23.0软件对LBP基因多态性位点与奶牛乳脂性状进行了关联分析。结果显示,在牛LBP基因中发现I2-1095 G>A、I5-142 T>C、I5-199 A>G 3个单核苷酸多态性位点,SNPs的多态信息含量(PIC)和遗传杂合度(H)均介于0.25~0.50之间,处于中度多态,经卡方值验证结果显示SNPs在群体中处于哈代温伯格平衡;经相关性分析发现,其中I5-142 T>C、I5-199 A>G 2个突变位点均与奶牛乳脂、乳糖、体细胞、校正奶呈显著相关(P<0.05),另外,I5-142 T>C位点还与产奶量呈显著相关(P<0.05),I5-1095G>A位点与乳糖性状显著相关(P<0.05);使用Haploview软件对SNPs进行连锁分析并结合单倍型与奶牛生产性状相关性分析,发现H1H2为最优单倍型。本研究结果可为中国荷斯坦奶牛分子选育、进一步提高乳品质性能等提供依据。
外文摘要:Lipopolysaccharide binding protein(LBP)gene is a member of lipid transfer protein family.It binds to cell surface receptors,initiates innate host response and participates in lipid metabolism.To reveal the correlation between LBP gene polymorphism and milk fat traits of Chinese Holstein dairy cows.This study took 133Chinese Holstein dairy cows as the research object,studied the polymorphism of the second and fifth intron segments of dairy cows by gene sequencing,and analyzed the correlation between LBP gene polymorphism and milk fat traits of dairy cows by SPSS 23.0software.The results showed that three single nucleotide polymorphisms i2-1095g>A,i5-142T>C and i5-199a>G were found in bovine LBP gene.The polymorphism information content(PIC)and genetic heterozygosity(H)of SNPs were between 0.25and 0.50,which were in moderate polymorphism.The results verified by chi-square showed that SNPs were in Hardy Weinberg equilibrium in the population.Correlation analysis showed that i5-142T>C and i5-199 a>G were significantly correlated with milk fat,lactose,somatic cells and corrected milk(P<0.05).In addition,i5-142T>C was significantly correlated with milk yield(P<0.05),and i5-1095g>A was significantly correlated with lactose status(P<0.05).Using Haploview software to analyze the linkage of SNPs,combined with the correlation analysis between haplotype and cow production traits,it was found that H1H2was the optimal haplotype.The results of this study can provide a basis for molecular breeding of Chinese Holstein dairy cows and further improving the quality and performance of dairy products.
